Yo I just won 3 medals in science * 4
    #16539660 -

I am officially a graduate and the old geezers at uni decided to give me three medals all gravitating around the common theme of me being an outstanding undergrad scientist :awegroove:

I never paid who knows how much attention during lectures but I was always trying to do perfect experiments in labs. A gel ladder runs crooked > throw out the whole gel. I messed up volume by 0.05% > start again from scratch. Roughly calculated it seems I've spent 300% over the allowed time limits in labs :shoosh:

Fuck it feels good to have no more uni on your head (at least until next fall).

Congrats what did you get your masters in?

I must not fear. Fear is the mind-killer. Fear is the little-death that brings total obliteration. I will face my fear. I will permit it to pass over me and through me. And when it has gone past I will turn the inner eye to see its path. Where the fear has gone there will be nothing. Only I will remain.

Its a bachelors degree in molecular biology. Going for doctoral studies next.

There were points where I forgot about drugs, food, sex pretty much anything other than doing my PCRs, westerns and whatnot. Its a pretty major obsession in my case.

Did you discover anything profound or get any papers published? Or was it for replicating pre-existing experiments?

There must have been something to go from B.S. directly to Doctorate skipping a Masters?

Can you tell us more about the experiments?

What are the names of the medals?

Congrats  bro! Your jimmies should be very unrustled.

Now make me one of these:

Re: Yo I just won 3 medals in science [Re: teknix]

I did discover something interesting in neuroscience but that was during the summer so it has nothing to do with the medals (I'm expecting to get the publication soon, the PI is kinda lazy so it might take a while).

As for the medals, one goes to me being boss and owning 100 people in first year

The other 2 are for my performance in 3rd year. Basically I didn't discover anything, I was just doing the dirty work. However, there was a big problem cause one of the markers I used to make my strains with was incompatible with mass spectrometry experiments. Basically I had to feed the cells some heavy nitrogen labelled amino acids but the marker provided the enzyme needed for endogenous synthesis of these amino acids so the engineered cells would not take up the heavy label. I proposed about 6 methods how to overcome the problem while my mates were lost in the dark. I solved a whole bunch of other problems and my supervisor really respected me for this. He offered me a PhD but I declined.

Cool, how did you determine it was endogenous synthesis that was the cause?

That's standard practice nowadays.  At least in the U.S.

...the whole experience is (and is as) a profound piece of knowledge.  It is an indellible experience; it is forever known.  I have known myself in a way I doubt I would have ever occurred except as it did.

I added N(3)-(1-carboxy-2-nitroethyl)arginine and the cells began incorporating heavy 15N. Then I deleted the marker once I had integrated my cassette and got consistent results.

Nice!

How did you know that would prevent endogenous synthesis of the marker?

Haha now I'm going to call you honorary Dr. Mellow from now on. Keep making incredible discovery's and continue asking tough questions, you are an inspirational scientist.

I just googled an inhibitor of the enzyme encoded by the marker. But there was another problem, the heavy arginine gets turned into glutamine, proline and lysine thus really contaminating the proteome with heavy isotopes so one must delete glutamine transaminase or something like that (need to check my thesis).

Good God, I can't wait to get into molecular biology classes. You're giving me a raging learner (like a boner except for learning) right now.
