Slants stallng out

Hey folks I've started several slants over the last few months and only a couple of them have fully colonized.  It's really strange a bunch of them started growing along fine and then just stopped like they ran into a wall.  It looks like this:







And another one has almost fully colonized except one area that has remained uncolonized:





Normally this would indicate contamination to me if it was on a plate but the uncolonized areas all look totally clean to me.  I don't see any bacteria and definitely no mold.  Even after months I've never seen any visual sign of contamination.  Has anyone seen behavior like this before?  It's like they just decide to stop growing one day.  They've all been stored at room temperature.  Most of them are doing it so whatever I'm doing wrong at least it's consistent.

Any ideas?

Probably out of air.  Are the lids loose and wrapped with parafilm?

Slants should be kept refrigerated.  Room temperature defeats the purpose of making a master culture slant.

Ah yeah it's probably air.  I missed the part where I'm supposed to leave the tops loose.  They have parafilm but the tops are on pretty tight.

I'm storing the finished slants in the fridge.  I just meant I keep them at room temperature while colonizing.  Once they are fully colonized do you screw the tops down tight?

Wait- I thought you wanted the air to be minimal with them, ie lids on.

Mine are in vacutainers (with lids) in the fridge.

Did they fully colonize with the tops screwed on tight?

Never screw the caps on tight with mycelium in them.  I leave the lids one full turn loose before wrapping with parafilm for gas exchange.
