mini BRF attempt- fighting with PE

Ive been struggling to get my PE off the ground.
In agar, its colonizes slow and weak looking.
In PF tek it contams before full colonisation.
So im going at it a new way unless someone advises against it.
Ive got these mini tablespoon size restaurant cups and im gonna do the smallest little inoculation in those things, then shred them and put them to WBS if they fully colonize.

Lemme know if you have an idea or advice.

Start on agar the right way.

How are you putting the spores on the agar?

(In SAB) via Syringe, dont have an inoculation loop.
1-2 drops per agar.
Ive inoculated upwards 7 different agars.
My 1month is at halfway colonization
6 of the 2 week olds (in mini rounds) have either no life or just started colonizing.

1 drop is already way too much spore solution for a petri dish in my opinion. Especially for a PE variety because there's less spores and more contamination per amount of spores than average or so it seems.

Make an inoculation loop. Put one drop on the loop then streak the loop. Otherwise you get germination everywhere

I usually do them at opposing sides to try and isolate.
I read your microscopy post so I can see what you're saying though.

So I can just make an inoculation loop? Guess Ive been living under a rock. Ill look into it.

PE is a finicky little devil.
