so you want a pure substrain... (PIC)
    #4332589 -

i'm at the 3rd generation in my petri experiment and i isolated this super-resistant super-fast super-rizomorphic substrain.
when i started the experiment i really had no idea about the actual purpose of a pure culture, i was doing it just for learning how to do it.
and now? there are many options:
1) transfer my super strain in a grain jar and see if it colonizes fast and fruits
2) make a LC with it (LC from agar! will it be a pure liquid culture or the dna will be messed up agin?)
3) keep it going for ever and ever for dozens of generations
4) ... (?)

Well, they say often when you think you got good petris, expose them to light and see which pin. Any substrain, even a beatuiful one, may refuse to pin, or pin crappily. Pinning on agar is usually a good indication. Then you can use either a peice of the mushroom or wedges from the agar to inoculate grain and a LC. The LC will be of that isolate (or clone if you do the mushroom tissue) and should be an excellent fruiter. Thats what I'd do

i will make another generation (4th) because it seems that there are still some sectors in this one, then i'll have it pinning in agar and transfer it to LC if they pin successfully.
just a last question: when i'll have fruits, their prints will be any better than the print i started this from?
i mean: is agar work something you do once for isolating your own strain and keep it going through multispore, or you need to keep the isolated mycelium alive for any time you want to use it?

You may want to bring that topic to advanced...I'm not SURE. I believe the spore prints will be more apt to be better flushers when they came from a cloned specimen, but still not as good as the cloned specimen. I think they tell you not to keep cloning generations though, degredation can happen I guess..so your supposed to alternate, clone one batch, spores from the next, clone from the next, so on and so on. Anno or agar would have info on it if you cant find anyone here or in advanced with a for sure answer for you.

by changing the substrate recipy from time to time, you will not get degen.

i did some transfers from peroxide to peroxide agar for about 7 generations
and still, no signs of any degen.

afaik, the spores from shrooms that were cloned are just as good as spores
from any multispore originated mushroom.

oh and the petri in the middle of the lower row looks the nicest

i have been changing the "food" for all generations:

1st gen.: dog food + cane sugar
2nd gen.: malt extract
3rd gen.(current): dog food + cane sugar + H2O2

next time i'll do malt extract + H2O2.
i will probably learn by experience about the actual differences between a multispore LC and a pure substrain LC... if there are any!

try switching back and forth with MEA and PDYA.

you might get some better results.

I make my own PDYA...here is the recipy.

-PDYA-
500ml water
5 grams of instant potato flakes(Potato Buds Brand)
9 grams agar agar
10 ml honey (Raw natural pure honey - Beemaid Brand)
1 gram of Nutritional Yeast

damn road, that is some super-nuted agar, a bit too much for my taste already.

BTW, if you show that recipy, ADD that only to pc it for 20 mins
in order to AVOID the caramelisation of the honey !

i will soon have some lab grade peptone and agar, i will probably use peptone alone, or peptone+honey, or peptone+malt.
beside that, my famous human sperm project is still in the plans. :penis:
any info / links about the use of peptone in micology? the friend who's giving it to me says it can be used alone, but he's not working on fungi and he's not sure about it.

I've read that if you continually isolate, you'll get to the point where it won't even fruit..

Is that correct?

I'll be doing Agar tomorrow...for the 1st time in awhile..

i've gotten some good "rizomorphic substrains" pretty much just luck on my first run with agar. how much light would be needed and how long would it take to pin an agar plate? I have some that have grown way out of control because I'm too busy to cut them up and use them. I've seen some of those pictures of a fruit growing from agar, how difficult is that? any potency from MEA?

You wouldnt get a dose off of 20 plates so potency doenst really matter...regular light, doesnt take long for pins to form. Its just normal mycelium.

pc for 20 to 30 minutes... better?

lolzz

this PDYA recipy works great.

many new strains have been started on MEA and this PDYA recipy.

Well, the amount of sugars in this is the same as in the standard PDYA (which uses only dextrose).
