I have searched the posts and could not find anything that takes into account the concentration level of mycelium in a liquid cultures. There are many great recipes and techniques here, but there comes a point when any LC will slow or stop expanding in the media its growing. At this point there is a portion of the nutrient mixture that is not going to be consumed. This becomes the separator solution when shaking/mixing the LC. MY THOUGHTS or QUESTIONS: 1. Some of the media will already be infused into the myco blob that is growing - IS ALL OF THE REMAINING NECESSARY OR BENEFICIAL TO INOCULATION? 2. If you pull out some of the unused media prior to shaking/mixing - WILL THE MYC STILL SEPARATE ENOUGH TO PULL THROUGH AN 18 GA SYRINGE? WILL THE INCREASED CONCENTRATION SPEED UP COLONIZATION? My suspicion is that having a greater concentration of myco that can pass through the syringe might accomplish a couple things. A. The nutrient solution will be used faster and the mycelium will search out new nutrient sources (Bulk Spawn or Cakes or whatever is being used). B. The decrease in moisture added to the bulk could help reduce opportunistic contam that was waiting for a little more water to start growing. C. It might decrease total colonization time for the Bulk or Cake. I am getting ready to run an experiment that will test this theory in a few ways over a few different Bulk set ups with some LC that i have that has just about stopped expanding. If you have any experience with this theory or any recomendations for possible vectors to address let me know.
