A 110q tub or a 66q tub. 110q is preferred. alternatively you can build one out of Plexiglas or something else like an aquarium but that's not the simple AF way. Some people have found tubs in the nearly 200q size as well, that will work too. you will also need Paper Towel, 70% alcohol (usually iso) gloves for your hands A torch (or anything that doesn't fuck around like those little alcohol lamp things) And some tools like a scalpel and inoculation loop(not pictured) and you will also need a spray bottle(not pictured), that's capable of making mist, with some warm water and a drop of dish soap (approx 500ml water for one drop of dawn or Palmolive) I color code my spray bottles Blue for mist water(for spraying substrates) Clear for 150ppm chlorine(not often used) Green for 70% ISO Yellow for water with a drop of soap for SAB spray 70% alcohol sanitizes better than 90+ or higher concentrations more info about that here do the holes with one of these get it hot on a stove, don't burn yourself. make the arm holes comfortably far apart so that you can move your hands around as if there were no SAB there in the first place. The idea is to have the same freedom of motion you would have if you spent a bunch of money on a flow hood make some holes, clean them up with some sandpaper if needed so they don't cut your forearms up. get a table and your SAB wipe the inside of the SAB and the table surface with some alcohol on a paper towel put the SAB on the table and you might want to put a weight on top so it doesn't move around at all put your work inside the box, (or put it on the table then put the SAB over it) spray the water inside the SAB with your sprayer on fine mist mode light your torch and put it on low, I put it on the side of my SAB on the table be prepared mentally for the work you're about to do have everything ready and rehearsed in your mind put the gloves on and wipe them down with iso, wipe your forearms down too WHY IT WORKS You CANNOT sterilize the inside of a SAB Sanitizers are not Sterilizers and Sanitary is not sufficiently contamination free to do STERILE work. thus you have to work with physics rather than your chemicals you do not want air movement, it's a STILL AIR BOX this is why glove boxes are a no-no in this hobby glove boxes with attached gloves create air currents, you don't want to stir up non sterile air, you want to avoid having the contamination end up in your work in the first place This design uses no sleeves or any caps on the arm holes. I believe my work speaks for the efficacy of not needing to over complicate things. now get to work!
