What % of nutrition in your mix? Across the board it just looks like a minor media resource imbalance of some kind but honestly not too bad. They do not look contaminated and they all look like the targeted species'. The semp plates look a little thin. The other cultures look a little thin and that ksss has a far reaching margin. I'm leaning on too heavy of nutrition and not stiff enough agar. Personally I would lower the nutrition, add a pinch of yeast, and up the agar powder just a hair. As far as the APE culture goes it looks like it has some internal struggle going on. One trick is to make an LC and bank some in a sealed container in the fridge for at least a week or two. Bring it out and plate some up and you'll see the culture "break" and sector really hard. Typically you get extremes where very rhizo sections run parallel to very tomentose sections. It's really easy to separate the more aggressive stuff at that point. I do this with clones and ms both if they are having trouble deciding what to be. I've seen clones break like that and they'll eject what are probably, and look like under the microscope, monokaryons. They were either useful, entertaining for the colony like a pet, or stowaways sapping the colony for resources. After a lengthy period of low metabolic action colonies take the chance to eject or discard those that are counterproductive to the colonies goals for whatever reason. The could just be damaged in some way genetically or whatever I can't speak for the desires and decisions of mycelium. This was just a phenomenon I noticed pulling cultures from cold storage over time so I started employing the occurrence on purpose to help with my own goals. These are just my crazy thoughts so take it as you will. Grain of salt and all that. ![]()
