Quote: I was replying to this post from PurpleHat about pouring plates and the agar separating. You LC legends are out of my league. https://www.shroomery.org/forum Quote: Respectfully, I suspect that you haven't foolishly let agar sit as liquid in a water bath for 12 to 20 hours. I don't recommend it, but do think that it provably will allow the agar to begin to separate (probably the carbs, too). If it only happened once or twice I would call it a fluke. But it is easily reproduced and also avoided by pouring in a more reasonable time line and with a long enough pc cycle. I fixed it after reading your suggestion to increase PC time for clearer plates. I believe that you have an Instant Pot from reading your Autoclave Validation and Load/Cycle Verification thread. If you are so inclined, sterilize a batch of typical recipe agar for 20 minutes (a commonly recommended but misguided amount of time ime), then leave it on the Keep Warm Low setting for 24 hours. You should be able to easily see the difference in clarity from top to bottom of a media bottle, when pouring, the top will be watery thin and the bottom will be sludgy thick. And at that stage, stirring and pouring without reheating to above 185f will not produce good plates. Not saying that this is everyone else's issue, but it was the cause and cure of mine. tldr; Thought I would save some time prepping agar the day before pouring and using the Instant Pot timer function or really long water baths always led to disappointment. Or at least inconsistent agar plate hardness. -------------------- Don't Panic Edited by ReverendMyc (12/07/23 01:26 PM)
