Quote: I only intend to keep "winning" isolates/clones in the fridge, so to speak. I only have 9 plates at the moment, and a bunch of empty prepped mini rounds. Agar is cheap even when you buy premixed premium shit. I've yet to see a plate contaminate at all. Pretty positive the first tub from agar spawn is a winner, too - given the aggressive mycelium and zero signs of contamination. I got lazy with one of my SAB sessions (long day at work, lighter ran out of fluid) and went about the rest of the tranfers in haste, even smacking the scalpel against the lid, etc - and I still didn't see any contamination. Current tub is just MS>Agar>LC, but will be good for taking clones which I also intend to put the "winners" on ice. I'm now into third transfers (fourth set of plates) - these very well may be isolated, I'll know in a few days or so. Anyone have any tips on visibility? So far I just hold mine to a 6500K flouro and try my best to see what I can. EDIT_ @ Pasty ~ This does make sense though, I hadn't thought of it that way - allowing the contaminants the opportunity to grow to see if they are there. The container I keep them in is basically a shoebox sized SAB without arm holes, I keep the lid cracked and I clean it out with 70% iso like once every 2-4 weeks. Ordinarily I just knock out any vector for possible contamination but in this instance it does make sense; especially for blended agar LI and such. Edited by plurfekt (05/10/18 05:27 AM)
