It doesn't take a lot of tissue to get a clone so maybe try to minimize how deeply you're dragging the blade through the stem? Maybe someone else has a better way though. I haven't seen any increased contamination from dragging the sample onto the agar.

A few questions, not so much related to your tek, it's pretty much cut and dry, more so just agar related. I took some tissue from a few nice fruits (one from outside that was 136g wet), how long until I start to see noticeable growth? Also, I was having the hardest time getting the tissue sample off of the blade and onto the agar where I wanted it, on a few occasions, I had to touch the blade and drag it to get the tissue off. My blade was flame sterilized, so would this cause any contam issues or is it proper technique to touch and drag like you would with an inoculation loop? Thanks for the tek, it's pretty awesome.
