Please see the photos of my latest experiment. Can you advise, if any of these dishes is looking good, or is contaminated?

I cut out part of the agar, where there was contamination, this is why there are holes in this agar.

Dishes look terrible. Definitely contaminated
Possible to get clean myc transferred, but in all honesty I would start over.

Ok, thanks. But how can I tell if they are contaminated, the dishes have only the black-coloured spores with white mycelium around them. I mean, it must be mycelium that develops around spores, right? I talso looks white and fluffy, like one would expect myc to be.

Please ignore the first pic from the left, I poured the agar myself for the first time and made it too thick and colourless, so it is hard to tell what is what. But the other dishes should clearly show black spores with white growth emanating from them.

Most everything from MS will be contaminated.
Just the way it goes.
This is the reason we do plate work. To get clean mycelium.
If you are good at plate work you can achieve clean myc in 2-4 transfers depending on cleanliness.
Keep transferring.
Work on your technique and form.

Thanks, do you recommend cutting off small bits with the most obvious spores with white growth on them and transferring them to a clean dish?

I think the reason for contamination is that I did not seal my dishes after transfers. I got some parafilm now and will seal them all. I have my own monotub- sterile box with holes for hands in them. I spray it down with 70% alcohol and flame the blade until it is hot.

Yes, I would recommend cutting off small bits of the outer most edge and then once you have a clean culture that is strong and aggressive put that to grain or LI/LC
The reason for contamination is more than that. And I’m suggesting that you take time to work on your technique either under a Bunsen Burner (blowtorch) or a SAB or FLOWHOOD/FFU
Also look into using cling wrap as an alternative to polyfilm.
Great option and long lasting with only a couple wraps.
Most spores prints or MSS come with contamination.

Thank you for your help. I have done that now, does this look better? I isolated the bit that I thought looked good and transplanted it. There are clumps in my agar, it is not contaminated. Also, the black spots in the transplanted bit are spores.
