spore syringe quality?

first, thanks to everyone on here for the amazing resources! first time grower, using agar tek from D3 and BOD, and planning to do Frank and BOD's mono tek using WBS once a i get the hang of agar!
ordered syringes from one of your sponsors, which i won't name yet, b/c this is prob my fault, not theirs. but also ordered some spore syringes of shiitake, lions mane, and blue oyster (for cooking!) and the content was so different i thought i would ask if I have bad spores to start with
first the B+ spore syringe from a sponsor:

just curious which is "normal"? the single clump of spores? or the murky swamp-like syringe?
here is my first two attempts at plating the B+... not sure if i actually got anything out of the syringe evey tho i shook it up a lot (it has been almost 3 weeks since i plated these) and not sure if this is mycelium or something vulgar... well one spot is obvious


Any feedback / help would be appreciated! Was pretty careful with my technique and used a SAB... but it was my first try.
Yesterday I ended up pushing all of the B+ syringe out into a sterile dish and grabbed the little black clumps with my flamed (and cooled) loop and smeared the suckers all over 3 new plates. will let you know how that goes...

The edibles are in liquid culture, the actives are spore syringes. 100% normal.
You wasted that syringe. You only need a drop of MS solution on a loop to streak a plate. Those black clumps are millions of spores all clustered together because you didn't shake the syringe well before use. It only takes 2 spores to start a culture.

thanks, i didn't realize the difference in liquid culture and spore syringes!
i did shake up the spore syringe each time, but the growth was so slow, i thought i was doing something wrong.
do the plates i posted look like they have some mycelium that i should transfer to new plates until i get something more rhizomorphic?

The top plate has mold at 11:00 and what looks like healthy mycelium at 1:00 growing in 3 different sectors. Bottom plate is too out of focus but it looks like I see a few sectors of healthy mycelium there. Both plates are ready to take transfers from.

The top pic looks like the good growth has fuzzy mold around it and nothing to work with IMO.
The bottom plate isn't very clearly visible but it looks like a good plate to grab some transfers from.

UPDATES! here are my best two plates that i unloaded a whole spore syringe on... 
i went out of town and let these grow for too long, i know, i know...
blue plates source

red plates source

I transferred 6 wedges to 6 new plates... going to give it a few more days to grow out... but what do you all think should be my new "paterfamilias"??

They need more transfers to clean up the cultures before you can try to figure out which ones to keep. I would have grabbed a transfer from 8 oclock on the blue plate source as the mycelium seems more aggressive there, but the others should work as well.
Red 3 is a goner IMO.

IMO yes - at least a T2 or T3 depending on how they grow out. I don't like sending random T1 growth to grain since so much downstream depends on the health of that initial culture.

woo hoo! this 3rd generation of plates seems to be looking pretty healthy, no? 2nd row, bottom right is what i was thinking would be my master going forward. thoughts?
 ready to spawn to grain?
growth has been pretty slow tho, this is 2.5 weeks in. i think it is b/c my house has been a little colder than usual due to the weather changes...

Those dont look right, it looks like it might be the plates though. Either too soft or the myc doesn’t like the dye.
You should be getting solid growth similar to this.

The germ plates look normal, so it’s either technique or the new plates.

ok well that's a bummer
maybe i'll go back to the previous generation and make new plates
these plates were made using D3's agar tek. but this latest set (purple) were made w a different batch of agar and different dye than the previous rounds (red and blue in the pics)
i'll just eliminate all dyes for now until i get the hang of it and go back to a previous generation

ok, so i took your advice, threw out the funky P2 plates, made new agar without food coloring, and transferred from the healthy red P1 plates a generation ago
growth is looking a lot more uniform
also switched to glass dishes... no more condensation, love it!
not seeing that beautiful rhizo growth, but do these look ready for grain? or keep re-plating the best ones until i get rhizo?

What's your recipe?
You can add 1-2g of nutritional yeast and use less malt extract (12-15g/liter) to improve culture appearance.
Make sure you're not overcooking it, 15 minutes at 15psi is plenty.
