How Long to PC One Cup of Agar
    #19533107 - 02/06/14 10:17 PM (9 years, 11 months ago)

So I just PCed a jar with 1 cup of agar for 30 minutes. Well, most of the agar was no longer in the jar! Duu!

The agar goes into my PC pretty hot to start so not to shock the jar and to help the agar mix. How long would you PC 1 cup of agar?

You should be PC'ing your agar media recipe for about 45 min.  If your new to agar prep, take a look at Fatguy's Agar Tek.

Yea, I read Fatguys Agar Tek. I just PCed another 1 cup amount for 10 minutes and again all the agar is out of the jar. The jar is only half full. What the heck is going on? 45 minutes? You've got to be kidding. Nothing will be left in the jar by then.

Is this because I am letting the pressure out over a 2 minute period?

RR says 30-40 minutes at 15PSI for agar media sterilization, but +/- 5 minutes wont make much of a difference.  The key to avoiding boil over is adding enough water to the PC that it covers half the bottle of your media, and it will cool slower and reduce the chance of boil over.

After the 25-45 minutes just turn of the heat and let cool for 2 hours, then remove the agar media from the PC. If you release the pressure immediately you will get boil over.

I was letting the pressure out to quickly I see. My seal on my MC is also old and not holding pressure once the pressure goes down a bit. Looks like a trip to Bed Bath and Beyond for a new seal.
