anti-biotics

I was wondering if you could use anti-biotics for indoor mushroom cultivation?I know that when mushrooms grow outside they develop antibodies to contaminants and they grow.But if u grow them say for ex. via pf tek, they don't develop these antibodies thus they have a greater possibility of contams.If u can use some kind of anti-biotic[s] is it practical?

for controlling bacteria with agar culture I add about 1/10 gram of gentamycin sulfate to each liter of media before sterilization Ive never used it in a PF tek but I suppose it could be done

theres plenty of antibiotics out there, i add ampicillin to my agar plates. i have also used getamycin sulfate. both work well.

Woot! that works?! I have a bottle of Gentamycin Sulfate, we use to mist our cats if they get an upper resp. infection.

We go to a vet supply store in my town. They have a liquid version of it and then they have a powdered version that is more bang for your buck, but you need a good digi scale to mesure it though. Its also used for cattle and such - so cattle feed stores might carry it also.

FYI, gentamycin sulfate can be added prior to sterilization, while ampicillin and many other anti-bacterials are heat sensitive and must be added after sterilization and some cooling...
make sure you read up on the info before you do it!

i reckon its often a waste of time to be honest. Unless youve got bacteria - which is USUALLY easy to isolate out anyway - my problem with wild prints is nearly always molds rather than bacteria. And if its strong bacteria the gentamycin is pretty useless....

Ok, I've mentioned this before, but been working on it a lot lately and this tek really works. I pick a lot of wild edibles up in the mountains and have been cloning/printing them for domestication. Often, both molds and bacteria are embedded in wild mushrooms. It's normally easier to transfer live mycelium away from molds than from bacteria, which seems to 'hitch-hike' on the myc.
If you'll mix up a batch of antibiotic agar, you can pour the hot agar directly on a petri dish with both mushroom myc and contaminants. Completely cover all the growth in the petri dish with a fresh layer of hot antibiotic agar. The hot agar will 'pasteurize' the contaminants, but the live myc will barely be set back. In a couple of days, the mycelium will grow through the layer of agar and appear on top. At this point, carefully 'shave' a bit of the myc off the top without taking any of the agar. Transfer this to a new dish and you now have a clean culture. The very dirtiest of wild specimens might require a repeat of the above, but this procedure usually gets it on the first go. Try it.

Sounds great, but at what temperature due you add the antiobiotics to the agar? I've heard that (with the exception of gentamycin), they will break down fairly rapidly at elevated temperatures. I always wait until the agar is about 50 Celsius (rule of thumb: cool enough to hold to your neck without burning yourself) before adding the antibiotics and quickly pouring.

PS--And to be quite technical, mushrooms do not produce antibodies. Those are a special family of proteins produced only by organisms with a specific type of immune system (e.g. mammals).

I use the antibiotic agar from fungi.com. It withstands the heat of the pc.

I like to use 10 mg of benlate (benomyl) per liter of agar to control molds. It is a fungicide that selectively inhibits sporulating molds while allowing mushroom mycelium to grow. I find it amazing that such a chemical exists and actually works. You really need a good scale though, if you add too much nothing at all will grow. Unfortunately, Dupont pulled it from the market and it is now very difficult to find. See also Carbendazim, the active metabolite of benomyl.
