I seem to be having problems with sterilization. Would adding a few heaping table spoons of salt to the water I boil my jars in be an effective means of decreacing the risk of contamination? It should raise the boiling temp of the water a bit (still gotta do the calulations to figure out how much). So, whatd'y' guys think?

No. Even if adding a few tablespoons of salt had a big effect on the boiling point of the solute(which it doesn?t, the effect is fairly small), you would only dry out your substrate this way. The boiling point of the water in your substrate stays the same, so you would be drying out your substrate because the temperature outside would be higher than the boiling point of the water in the jars. You would achive the same effect if you would stick the jars into the oven and turn the oven higher than 212?F.
How long have you been boiling the jars? Which substrate? What containation did you experience?

Oh yeah! I haven't thought of that. I've only tried growing once. All the jars got contaminated. I think I boiled them for 70-75 minutes. Come to think of it, it probably wasn't the initial sterilization that wasn't up to snuff, but the practice of sterile procedures during incubation. I didn't tape the holes and didn't serilize the area before examining the jars. I used your "PF Tek for Dummies" thing. What's your opinion on the ramsey seal?

I tried it once, failed to see any benefit, abandoned it therafter.
You used the PF-Tek for Simple Minds? On sterilisation: ....you can also sterilize the jars using a big pot with a lid. In this case steam the jars for 1.5 hours in a pot lid on.....

yeah, I felt that the jars would get to dry if I boiled them for 1.5 hours, and they did. I tried to salvage what uncontaminated mycellium I could before deciding to order another syringe. I used a chunk of colonized cake in some sterile honey water, it sort of worked. Anyway, when I opened the jar it was really dry and shrunken. I guess you learn from experience, next time i'll keep the incubator more humid. I think I just need to be more aware of the fact that everything needs to be sterile when dealing with this sort of stuff. And what happened to your web site?! Did you move it or something?

if you are following the pf-tek or the pf-tek for simple minds there are some things that you need to know first. a couple of the things they say are wrong. first of all, they tell you to incubate your jars at 70*. this will work but will take forever, optimal temp. for incubation is about 85*.
they also didn't tell you that your temp has to drop by 10* to initiate pinning, so you should be incubating at least 10* higher than the room temp that you'll be fruiting them in.
i almost made both of those mistakes cuz i was following the pf-tek word for word, but them someone told me that 70 was way too low for incubation.

man you only have yourself to blame for contams i have karo jars that have the slowest growth in history..been well over 2 weeks and they still haven't contamed..first time ever..my wbs jars no contams at all..lysol oust and place your jars in agallon freezer bag then just inject through the bag..which i lysol the inside of the bag..so i have like a mini glove bag..

Oh yeah, another thing they say in the pftek is to put give your jars indirect sunlight during incubation, this is WRONG. they should be in complete darkness. i dunno why it says to incubate them at 70 degrees in indirect lighting, but the faq says to incubate them at 86 degrees in darkness until it's time to initiate pinning.
