Panellus stipticus
    #2060470 - 10/31/03 12:14 PM (20 years, 5 months ago)

Hey all, my garden finally started putting out a few Panellus clusters.  When they were young, the glowing could be seen even with some street lighting nearby.  The grain jars are still glowing like mad.  Despite this, the damn things will not show up on my digital camera, even with maximum aperture and several seconds exposure time.  I really don't know what I'm doing photographically, so there could be some operator error.  Anyway, the glowing of this cluster was beginning to weaken. . .it had been on the log for a couple of weeks.  I cut it off, took it to work, and used a chemiluminescent imager that we use for incredibly weak signals from things like western blots.  It's not a cool color picture like I've seen before, but I thought it was pretty cool.    The imaging box is specifically designed to be absolutely light-proof, everything you see in the black and white photo is coming from the fungus

I have another younger cluster that is coming up.  I'm going to get an image of that one, along with a year-old grain jar that is still glowing like crazy [and pinning, surprisingly].

Happy Halloween!

Nothing really special, alder logs plugged heavily with dowels, buried. . .
Only 2-3 of these logs were Panellus, the others are reishi and P. pulm. . .

I'm pretty sure the culture was from you, armedia.

Cool. Now that I have my garden settled, I'll plug some oak logs next spring.

As for your year old jar: six months is my record. I have a pet theory that the H2O2 I always use in my cultures, as it breaks down, provides additional O2 to the redox reaction that is the source of the bioluminescence, allowing maximal brightness.

Glad to hear my culture is spreading far and wide.

People were looking at me wierd when they saw me sticking a jar in the imager

This champion of a jar is just under ONE YEAR old.  The pins are a couple of months old.  If you can get someone to shut themselves in your closet, these Panellus jars are great to share with myco-newbies.  Oh, and I was wrong about it being a grain jar, it's actually mostly dowels [innoculated with birdseed].

Here's the 'more fresh' cluster.  You can see how this one was much brighter, it was even illuminating the paper towel it was sitting on.

For anyone interested in more information about the bioluminescence pathway, this is a short paper on the subject:

http://www.ecol.kvl.dk/~sto/PDFposters/OlssonRedox2000.PDF

The practical thing I took from this paper is to let the jar incubate fairly well-sealed for a few days, then loosen the lids before 'viewing'. It really works, and only takes a couple of minutes for the increased luminescence to occur. The same principle holds for agar cultures. . .keep sealed in parafilm until ready to 'view', then remove the film.
