Jars too wet/Substrate too wet - Rye - HELP
    #628087 - 05/12/02 07:13 PM (19 years, 6 months ago)

I innoculated a number of rye jars with liquid substrate (honey water) and let them sit for a few days. After the weekend they started growing mycelium and have since been going strong, but it seems that at the bottom of the jars the honey water collected and made some of hte rye grains explode. Now a number of the jars are quite greasy and overly wet. I remember reading somewhere about using activated charcoal to cancel out the wetness. I see the FAQ suggests adding some gypsum to help.

Does anyone have any thoughts? Any help is greatly appreciated.

well here we go again. the prep of your rye is wrong causeing the problem coupled with honey water. Well I guess the honey water we will start with cause thats where you started. For liquid innoculation I rather use a eber blender with an agar dish. Blend up and all you need is a drop to get a culture going. Im not sure about your honey water but when my fire ants try it the mycelium likes to stay seperate from the liquid theerfore you need to innoculate with more fluid to make sure some mycelium comes out. This is where too much water came from. Also bacterial could have already been inside your honey water. So lets get rid of the honey water. All you have to do is transfer some mycelium to get another culture going. Have any colonized brf half pints? These work all right for colonizing pints of grain. In a glove box/flow hood scrape a bunch of small chunks of your pf jar and add to grain. There you are done but I dont like mixing teks I rather do a faster easier grain to grain transfer. Take one pint jar of grain use a fork and lossen the top 1/3 or 1/4 layer and dump into a quart jar. This is faster cause the little colonized kernals can be moved throughout the jar causeing extremely fast colonizing. Faster the colonization the less the chance of contams. So we covered eliminating your honey water and proper spawning of grain now lets talk about how you preped your grain. Rye is a little difficult to work at first but once you understand getting the right moisture content in the grain it is easier(less work, no cooking) then wheat. The only method that has ever worked for my fireants after going through more rye then you weigh was a overnight presoak. Easy enough there are no exact measurments needed for this tek because its all about the mooisture content of the grain. Get a bucket pour your grain up half way add a bottle of h2o2 (not necessary but is always done because we feel it helps) and fill the bucket to the top with water. Cover with aluminum foil and let sit overnight. To tell if grain is done it should be chewy and slightly moist in the center. Grain may smell at this point h2o2 helps keep that odor down also. Now dump all the water off of the grain or strain good. Load the jars up and you are now ready to go. If rye sprouts it is ok h2o2 also keeps sprouts down. I never use vermiculite in rye jars only wheat. Good Luck!!!!! Actually wish me luck Im going to go apply for a job here in 15 min.

i use rye grain all the time and all i do is put the grain in a pot, about 1/3 full, then add water till it covers the grain plus another inch or two deep. bring to a soft simmer and stir occasionally. you will know when it has absorbed as much water as possible when you see a few of the grains explode. then, immediately turn off the stove and put into a collander to drain all the water out. after that, i put 3 coffee filters instead of the metal top ( with a 2 piece top using the metal ring to hold them on) trim the excess coffee filter off, cover with aluminum foil and pressure cook at 15psi for an hour. i use quart jars and havent lost one yet. i innoculate through the filter, then put a tiny drop of superglue there and accelerate it.put the aluminum foil back on and incubate.

maybe the coffe filters help to dry your substrate out. I have never had success with steeped rye, always too much moisture especially when it is saturated enough for kernals to explode. What is your contam ratio with coffee filters? I always heard they didnt work so good. Luckily enough I live near a tyvek factory always wondered what they made there until anno started mentioning tyvek.
