Mushroom clonning... * 1
    #949722 - 10/10/02 01:16 PM (21 years, 6 months ago)

Hello !
I have tried to clone an oyster mushroom (from the grocery store). I have 6 agar  jars (instead of petri dishes) with 6 little pieces of mushroom. This is the 4th day now, and the little pieces of mushroom have "furr" on them... Is it normal ?!

It could well be mycellium....or it could infact me a contam. Welcome to the fun

First off, what oyster strain are you trying to clone? Did you use aseptic conditions?

I'll dig out the mycellium characteristics for whatever oyster strain and post them for you.

I don't know what strain it is   All I know is that I've bought a pleurotus ostreatus...
(Well, I think so)

This is what it looks like ( the white fur and there is also a contam there) Do you think it might be mycelium ? All my 6 jars have that. I've used a glove box, but this is my first time...

Isolate the white mycelium from the contamination ASAP.

It looks good! The white stuff in the center is mycellium. Just cut a piece that is as far as possible from contamn and transfer it to a clean jar.

It looks like nice rhizomorphic growth, although the pic is a little blury. Make sure to steralize your instruments (scalpel, whatever) before doing the surgery to remove the contam!

Keep going....don't let a little bit of contamination put you off

The golden Stametsian rule: You should allways remove the healthy growth from contamination, never the other way round.

Anno is right. You have a far more greatter risk of recontaming the culture if you try to remove the contam, due to you might spread more contam spores all over the culture, or by not getting it all. So it is much easier to just take a good part of the contamed cuture and put it on a new sterile agar dish. If it still contams well best to start over. Good luck................peace

Hey guys ! I followed your advices and everything seems fine !
I have another question... When do I know that the agar has been completly colonized ? When the mycelium will cover all the agar ?

Correct. Or, if you're not so lucky, it'll go invitro on you ;-)

mycellium will only colonize the top surface of the agar.
dont expect it to colonize the sides and/or bottom of the jars.

...and furthermore, the usual advice is to transfer it before it even reaches the sides of the dish (or jar)....

Jars... hehehe I messed up with the petri dishes; I just don't like them. Anyways, I believe I have a fully colonised jar now  and I've never been happier in my life!  :smirk:

Well, I have extented my cultures to almost 20 jars now. I've also made a incubator, so I've put my cultures there and they are growing faster!
The culture in the pic is still growing, even if I had to cut some parts of it !! The mycelium is growing vertically!!!  :ooo:
I have two jars of grain right now, and I'll inoculate them tomorrow!

I think I have some problems with my cultures... the agar is turning to a dark yellow and mycelium stops growing... why is that so???

By the way, I have an incubator and the temperature inside is +/- 75
Could the heat be a problem ??
