Well hopefully someone else sees the oddness in trying to prevent your brain from having maximal serotonin availability... in order to have a cheaper dmt high.
the synaptic cleft has to do with the downregulation of 5ht receptors.. i get all this. The point is that the serotonin is being released from an axon, after being generated within the cell nucleus far away.
The extra serotonin is being released in an orderly fashion according to the voltage potentials across the axon do you disagree?

no but again, it's a function of ligand (substrate) concentration dictating signal transduction. that's the whole point of this thread.
why would you want to maximize serotonin concentration? would you really want serotonin syndrome?

I'm still curious why 5-HTP significantly potentiates cannabis highs...

that's a good question, I have no idea. cannabinoids are such a complex ligands they influence a wide array of cell signalling (acetylcholine, dopamine etc.)...and of course these signalling pathways influence other neurotransmission downstream, and vice-versa.
couldn't tell you about 5-htp, but ime, DMAE restored my short-term mem, and sometimes killed the buzz from weed.

umm maybe you should read this:
http://www.ncbi.nlm.nih.gov/pubmed/16023217?ordinalpos=1&itool=EntrezSystem2.PEntrez.Pubmed.Pubmed_ResultsPanel.Pubmed_DiscoveryPanel.Pubmed_Discovery_RA&linkpos=2&log$=relatedreviews&logdbfrom=pubmed
the conversion of LT to 5HTP is the rate limiting step in the production of serotonin, it's like the law of mass action y'all
still the brain is a complex thing
just cause there's more serotonin in the cells doesn't mean it's going to be regulated. vessicle release is a highly regulated process

that's a good question. I couldn't find anything useful and short search inspired by this question, but I did find that cannabidiol can bind to 5-HT1A receptors, and some of psilocybin's effects have been thought to be due to 5-ht1a receptors. so that's a little interesting anyway.
also 5htp reduces spatial memory reduction from thc in rats - as well as 5-meo-dmt!

I'm not trying to jack the thread but I've been taking 100mg of 5-HTP every day for a little over a month now while smoking pot daily and I've never noticed any difference in potency with cannabis. Could just be me.
I do enjoy the effects of 5-HTP though. It seems to make me real light headed and fuzzy for most of the day. And my mood has definitely changed. I wouldn't worry too much about it being a dietary supplement as doesn't really suppress my appetite.
Just my two cents.

yes hello everyone out there to all psychedelic warriors, I'm kind of new to this board and hope I'm posting in the right place, but I have a question, a worry, a concern and hope I can find some answers. I recently smoked 5-meo-DMT for the first (and probably my last) time. While the experience was mind-shattering and in the end, overwhelmingly positive, some not so positive things have started to creep in. It started two days ago when I was on the train (to my parents house of all places) when I suddenly felt like I was "launching" again. My hands felt numb and nothing was real and I started into a full-on panic attack. After a few hours (!) it was kind of all done, but in the days after the major attack, I still feel like it's there. like I'm tripping a little bit all the time. Rescue Remedy and Kava seem to help but I'm kind of freaked. I just took 50mg of 5-HTP and hoping for the best. anyone out there have similar experiences? I'm starting to feel like I'm in "Jacob's Ladder" or something. I want my brain back! please, and thanks.
