Quick questions RE: Practicalities of Agar pouring
    #945429 - 10/09/02 12:48 PM (14 years, 1 month ago)

Sorry about this one, but i'm just honing my techniques and trying to tie up a few loose ends.

1. Do you parafilm the dishes right after pouring or after you innoculate? If so, doesn't that pose a problem when innoculating, since parafilm is self-sealing?

2. When pouring multiple plates is it better to wrap dishes individually as they are poured, or is it better to wrap them individually after all have been poured?

3. How much of a concern is condensation? If not a hinderence to mycelial growth, how much does it impair viewing of the culture? Is there any way to eliminate or minimize condensation? is it advisable to wipe away moisture with a sterile cloth, or is that just inviting contamination?

I am uning a flowhood which is 2'x4' and is 99.97% @ 0.3u that has HEPA-esque furnace prefilters.

All helpful replies are welcome and greatly appreciated!

Thanks again,
H

In reply to:

I seal the plates after I innoculate. Trying to wrap a dish when the agar is still fluid sounds very messy to me.

I pour them all at once. I feel this helps minimize exposure time and allows you to be quicker and more efficient.

In reply to:

I don't worry too much about it.

In reply to:

I find that if I let the petris cool while stacked on top of each other, it greatly minimizes condensation.

In reply to:

is it advisable to wipe away moisture with a sterile cloth

No.

Good luck

Thanks for the input! Don't you run in contam problems by not sealing until the plates are innoculated?

Thanks again, input from others would be greatly appreciated as well.

Thanks Anno, i was hoping you'd share your thoughts! Just a small addition to the list above:

Do i need to sterilize the Parafilm? i'm assuming this would be done by wiping it down with alcohol, but ive not heard of this before, so im assuming its unnessesary.
