vaccume seal after PCing?

After Jars were pressure cooked, in a smaller Pressure Cooker, they were take out still quite hot and the lids were screwed on tight. These were then placed in the fridge for cooling...

Any suggestions for innoculation? Will there be a vaccume type effect that will suck airborn contams into the jars? The lids are flipped, rubber seal up, and taped to the ring. There is also no dry verm layer to prevent contams as the jars are grain and being used as spawn.

If you have nothing to filter the air entering the jars with they are likely to contaminate; if not now then later. I suggest if you are going to innoculate these that you use a substantial amount of liquid mycelium or a big piece of agar culture. The mycelium could outrun any competitive molds or spores if there is enough of it. Invest in some polyfil or some filter discs. At least put some coffee filters over the jars if you don't have either of the above on hand.

He said that he had taped the lid to the ring, so they should not contaminate.

I agree.


If your are innoculating with a spore syring you should be fine. just make sure to inject without unscrewing the cap.

If you are innoculating with agar or liquid mycilium you will have to take off the cap and probably introduce contams. To combat these I would recommend injecting the jars with 5-10ccs of hydogen peroxide.

If you plan to innoculate with a spore print you'll have a high chance for contams, as you will have to open the lid, but can not use h2o2 as it will kill the spores.

Let the jars cool in the pressure cooker, and don?t seal them.

I have to go with Anno on this. Let the jars cool in the PC and you will not have problems. By cooling the jars fast you create a vacuum. From what I have seen the more you try to rush the process the longer it ends up taking. By trying to speed the process you just create delays for yourself.
