I believe that syringes are the one case where distilled H2O is recommended.

...then there is the long-term storage tek.
...and I used it with my dehydrator, to prolong it's life.
Well, tap is fine for soaking grains, making cakes, etc.

thanks, i thought that would be the case, i bought sterile 10ml syringes so do i just boil the water, draw up and expel a few times then do the final draw, wipe needle with rubbing alcohol towel, put needle cap back on and then into baggie to cool for a few hours?

Well, I built a flow-hood and skipped making syringes.
...but one approach that seemed smart to me, that I think Doc recommended, was to put the syringes, full, into a quart jar and PC them. You can put a jar of water in the PC too, if a GLC is the goal.
...but your scheme sounded good too.

thanks i do dont have a pc, you think i should do what was going to do but put water filled syringes into boiling water for a while?

boiling water and flushing it out a few times then letting it cool in a baggie works fine.

Iv used well water myself before  It would prob be best  Just be sure to boil for 8+ mins

do i need to flame sterilize once i have drawn spore solution back up the syringe, or will wiping with alcohol be sufficient?oh and how many 10ml syringes will i get from a +- 20mm print?i think its that size ,i got them from a vendor b+ and mazatec

flame after every jar and dont use alcohol to wipe your needle after its sterilized by fire, nothing is more sterilized than fire. wipe the injection sites b4 you inoculate then flame sterilize and inoculate. 
and you should get at least 6 s off a print that size .
it only takes the size of a #2 pencil eraser head to make 1 - 10ml/cc

i dont know what water would be better for making my substrate, will the water from a well 800ft down(i drink this water everyday)be better than normal tap water(also reasonable quality)?. the thing is i see a little green algae or mold on the spout of the well tap, this makes me think of trich, or will sterilization of jars kill any potential contaminants?

The water is not a critical issue, unless you are making syringes. Sterilization will handle whatever it adds.

thank you, and another question, if i make up and sterilize jars now(6pm)could i let them cool over night and inoculate in say 24hrs? or is that to long and i should just sterilize the jars a few hours before inoculation the reason i ask is because i am about to make my syringes and then will have to leave them for 24hrs to rehydrate.

I agree. I have 9 quarts powering down in the PC right now.
I will not be able to inoculate them* until I get back from a scouting event with my wife and son.
Seeing as I expect them to stay clean for the couple of weeks that it will take for them to be fully colonized, I am sure that Sunday will do.

I have jars of these three already going, but they are very cool, and I am sure that I will want to do a few more experiments with them.

just made seven syringes, i hope that all went well, i am going to make up substrate now, load jars, sterilize in pot for 90 min, let cool over night and inoculate some time tomorrow as i wont get time during the week.
