Two simple questions, fleeting thoughts that passed through my mind.

Has anyone ever tried to innoculate in test tubes? (i.e. the petri dish concept I'm seeing alot of)

Has anyone ever tried innoculating the same substrate with two or more different kinds of spores? Any theories or experiments that demonstrate if the different strains' mycellium would compete, or would it co-habitate?

These are both well known techniques. You could probably find more info about the test tubes in the agar faq, they are called slants. A search of past posts should turn up info on multi-strain innoculation.

Multi-strain innoculation basically does not work, one strain will dominate.
If you want to make a hybrid of 2 strains then you have to mate mycelia from individual spores (monokaryotic). Needless to say it is very difficult to isolate single spores. Plus the hybrid generally has less desirable traits than the parent strains.
