Sterilization with Ethanol?
    #926909 - 10/03/02 01:46 AM (18 years, 11 months ago)

I was wondering if anyone had any thoughts on the viability of sterilizing a growth medium of vermiculite/flour with ethanol. Seems like it should be even more effective/reliable than boiling, and possibly less of a hassle.

It's just not practical. Get a pressure cooker, then you can sterilize just about anything you'd need to.

Sure ethanol will kill anything in 10 minutes, but how are you going to get it out of your substrate? How are you going to guarantee it comes into contact with all of your substrate?
People have considered this problem for decades and the only satisfactory solution is heat.

maybe instead you could use a mix of 50/50.. h2o and h2oh...instead of just h2o...because the h2o2 will kill bacteria and after that it turns into h2o..so just wait a day before innoculating...a friend did it and it has worked so far with no contams

yeeeeeesss
we've all been heat sterilising substrates for years on end, just because we didn't have the benefit of your supa-intelligence. Why didn't we think of that before?!

psyphon - why is it not practical? it may be more practical for some than pressure cooking, everyone has different access to different resources.

zeta - but how are you going to get it out of your substrate?
the boiling point of ethanol is extremely low, as long as there's a decent amount of surface aread it should definately be gone in ~1day

-How are you going to guarantee it comes into contact with all of your substrate?Hrm, seems like a potential difficulty, but I'd bet it would do fine if you mixed it up, and had a sufficent quantity of ethanol. Or at least, I see no reason why it wouldn't

Champion - don't be an asshole, I searched for info on this and couldn't find any. I have several reasons why this would be preferable for a small batch, and so far zeta is the only one that has raised any potential issues, and i don't think they would really be a problem

1) Ethanol does not kill everything.
2) How do you intend for such a large amount of Ethanol to evaporate in a nearly air tight container?
3) How will sterile water get into the substrate?
4) What are these reasons you have for prefering this to standard head/pressure steralization?

isn't ethanol dangerous and hard to find outside of a laboratory? i think you have to have written permission to get it in the labs here.
