My Kitchen Smells like PC'ed Grain!
    #9218522 - 11/10/08 04:58 PM (12 years, 3 months ago)

I am going to be experimenting and sharing.

I will be doing my fist attempt with a Print as opposed to a syringe.

But, I am prepared with glove box this time!

It's a little Koh Samui print.

he means he's putting spores from print straight into grain jars. A risky maneuver for contaminants.

That is what I will likely do..

Without contaminating  the print hopefully.

I am likely to attempt my first agar work too.

good call that's supposed to be the best way to isolate a good strain without senescence. Just watch your jars every day for contaminants and plan ahead in case something goes wrong spores take a while to germinate and colonize the substrate. This is why we prefer to inject from spore syringe rather than to open the lid and drop in spores.

If you've prepared your foil print under strict sanitary conditions and everything was sterilized. I.e, exacto knife/scalpel/inoculation loop and kept in a sterile environment prior to opening inside you glovebox.....foiled wrapped...

and I'm talking about your agar jars too prior to pouring them keep them wrapped in foil until you open them in the glovebox....

Anyways, back on topic here I am so stoned I'm all over the place with this... please bare with me.

Just use your loop/blade and scrape a small portion of spores onto the blade/loop and drop them in....It works just fine as long as you keep everything clean, no contact with any outside air/hands, just gloves after pc'ing it

Never failed, from there I did g2g's...my pic's have 4th gen g2g transfers, the original masters were inoculated with a foil print I prepared in a baggy similar to the ones in the pictures... worked fine for me dude, and your print is pretty dark so you should be able to see visible dust on your blade, not like a really light print thats very hard to see spores on unless you have a microscope or something.

Don't mean to sound like I'm bragging, just giving you the basic fundamentals of inoculation.
