sorry if this is the wrong forum but this topic is what i would use this for.

but any ways my friend today brought over this microscope and we were wondering how decent it would be for identifying spore its a nikon alphaphot ys2 if you need anymore info i got it right here next too me

lol it has 2 eye thingies it has 4 10 40 and 100 it uses artifical light i put a little scrap of weed under and i could see the deep down tissue it looked liked

You need 400x to view spores & 1000x oil immersion to examine spore ornamentation.

As far as I know ToxicMan & Mr_Mushrooms use microscope. Check their posts, like this one:

If the highest magnification is only 100x it might not be very useful for looking at spores for precise measurement or examining ornamentation.

It sounds like it uses direct illumination (light from above the specimen, rather than through it from below). At 100x it should give some pretty nice views of gills - you should be able to see the spores, basidia, and cystidia pretty clearly. For some mushrooms that could be pretty helpful for identification. Also, even though the magnification sounds too low for measurement of spores, you should be able to at least judge the approximate size and shapes of the spores, which should be helpful. With a little practice you should be able to get within a couple microns of the spore size.

One observation you should be able to make with it that is helpful is seeing how many spores there are per basidium. The common store mushroom (Agaricus bisporus) gets its scientific name from the fact that most of its basidia have only 2 spores, not the 4 most basidia have. The same thing is the difference between Amanita bisporigera (2 spores) and Amanita virosa (4 spores).

I would say that it could be a useful tool for identification. The lack of the ability to view spores at really high magification makes it a bit less useful than a standard microscope, but you should be able to learn a lot from using it. The stuff I learned about microscope usage is based on using a high magnification microscope. I haven't seen any books on using a lower powered microscope - try looking and see if something has been published.

Try holding a digital camera up to the eyepiece and snapping some photos that way. That's how I do mine, and it works better than you might first guess. If you've got binocular eyepieces through two objective lenses you might even be able to do some stereograms, which would be great.
