I'm about to order some agar mix.

Is there agar that is the "Swiss Army Knife" of agars??

I'm thinking of getting potato dextrose agar.

Is there an agar that will support all fungi??

I'm sorry if this has been asked before!

i believe you add nutrients to the agar. the agar is just there to solidify the whole mixture.

Well. Would somebody be able to culture any fungus with Potato Dextrose nutrients for instance?

I can't promise any fungus will grow on PDA but I'm pretty sure any active mushrooms will.

A rule of thumb is to not always use the same mixture when you transfer to keep "challenging" the mushrooms to create a variety of enzymes.  If there is a brewer's supply or wine making store near by you should have no problem getting dextrose and light malt extract for MUCH cheaper than you'll pay for it in the pre-mixed agar powder.  If you buy just strait up agar powder you can make many different recipes and you wont be paying for the cheaper ingredients that make up 50% of the mix anyways.

Buy agar agar powder and add your own nutes is my recommendation.

I mean. If the mycelium grows then isn't that all it will do regardless of transfer? I mean. If I grow cube strain on PDA then transfer to PDA again won't it be the same as if I transferred it to a different nutrient the second time??

I'm kind of confused!

Okay, a few useful facts.

Mushrooms digest their food I think this is pretty well known.  Enzymes are used to break down the nutrients into usable sources of energy.  Over time if the mycelium is living solely off the PDA mix it will lose it's ability to digest new food sources that will be found in your grain spawn and bulk substrate.  This will lead to a lack of vigor known as senescence a problem when trying to store a master culture.



Wasting money is inversely proportional to the amount of research done.

So if the food source remains the same it will be fine with that source?

For instance. An outdoor bed of P. Azurescens would continually thrive from the hardwood, but if you had a P. Azurescens culture stored in PDA then eventually it wouldn't be able to consume hardwoods?

So which nutrients do you all use to diversify your master cultures' diets? Can you use only two and switch them back and forth? How long does it take (approximately) to render a strain sterile, or dead, for lack of a better term?

Unless you're going to get into mushrooms that aren't regularly discussed on these forums, MEA or PDA will work fine for everything.  All of the edibles and actives I've seen discussed here work fine on these two.  If you have a specific species that you're concerned about, post it and we'll see if it -might- just have a different requirement.

O.K.

Now I'm kind of interested in how this works. I just read the Wikipedia article about senescence but I'm still a little cloudy.

So if a species, for instance Ps. cyanescens was grown on PDA continually. It would not be able to absorb other sources of nutrients eventually. For instance MEA. Right??

I wonder why this happens. Well the enzyme production. I find it odd how this happens.

Malt Extract Agar (3%)(MEA)(General Microbiology)(FDA M93)

    30 g Malt extract
    20 g Agar
    1 L Distilled water

    Boil to dissolve ingredients. Avoid overheating, which causes softening of agar and darkening of medium color. Autoclave 15 min at 121°C. Dispense 20-25 ml into sterile 15 x 100 mm petri dishes. Final pH, 5.5 ± 0.2.

    This medium is recommended as a general maintenance medium. [1]

All senescence means is is "getting old"

All living organisms have a natural lifecycle and near the end of it they become weak and don't work properly (as you will in your late 60s 70s)

Cube mycelium will start showing signs of ageing after 3 months of life. Ideally it should have fruited by then.

This is why it is only recommended to do a G2G three times before fruiting for example.

3 weeks to colonize the grain from spores, 2 weeks each for two more transfers, 2 weeks to colonize your bulk substrate and then you have 3 weeks margin to fruit.

This is also the reason we keep master cultures. If you kept growing out the mycelium from the older sample it would age way to much. By being able to go back to your 3 week old master slant which is kept in a dormant state and growing out from that, you ensure that your mycelium does not present senescence.

It has nothing to do with the kind of food it has.

The only thing you might notice by not switching media is that for the first few days there is a slowdown while the cells activate the DNA responsible for the required enzymes and actually start producing those enzymes.
