okay its day 11 my jars were at 72 degrees F for 6 days, and now they have been sitting at 82 for 5 days and still are. Out of 5 jars only 1 got a fuzzy white spot 3 days ago, but the spot has not grown at all and is as big as a pin head. I used the PF-Tek With Amazonian spores, and 2cc per jar with no tape over the holes.(has dry verm layer). There seems to be no growth at all going on. Can a 40 watt incandescent sun bulb in the middle of the room on the roof kill the spores? Why is there no growth on the fuzzy white spot in 3 days?

It takes time to germinate. It took my grain jar nearly 3 weeks to show mycelium let alone a couple weeks to fully colonize. Unless using agar, LC, or g2g it will take a while to fully colonize. Multispore syringes are by far the slowest.

do the cakes colonize from the outside in or the inside out? I would think from the outside in or wherever inject first right? also should I see a noticeable difference in growth in the small white spot in three days thanks

There are some factors that play a role in how slow your multispore inoculation germinates. Sure they are not going to be as fast as a LC, but you also have to take into account the "Viability" of those spores being used. How old are they? How were they stored prior to you getting your hands on them? I've had jars take two weeks to germinate and then I've had jars take 3 days to germinate. I've even had a multispore inoculation go from inoculation to fully colonized in 8 days.

It all depends on where you injected. I injected dead center, wish I didn't. Also if you shake up the spores it will take longer for them to find their pair to germinate. As for the growth it all depends. It took a while for me to get to 30% colonized but once I shook it the jars took off, one is still slow though. It is hard to give an estimated time, you really just have to forget about them, patience

I knocked up 24 jars of wbs. with 4 different strains. The amazon spores are definitly taking the longest to germinate. room temp for me is 79F. amazon spores take like a extra week or two for me to see signs. Im sure you will see the myc soon. Be patient.

80 is what you want it will normally take twice as long at low 70's.

Higher the temp the higher the contam rate though. I would rather wait 4 days and have 10 jars instead of having it 4 days earlier and have fewer jars, in my opinion. I like the idea of a large tub with a heater blowing directly on it. Keep it at a good dry temp, less contams.
