*THERE WILL BE AN UPDATE RIGHT BEFORE HARVEST, AND HOPEFULLY WHEN MY CUBENSIS START PINNING, WISH MY LUCK ON THOSE CUBES!*

Introduction

So after one of my friends tried growing, and had some success I decided to take my own shot at this, and one of the main reasons why I'm doing this grow log is to keep him updated about my grow, but also share it with the shroomery community, and hope for some comments and advice. I started lurking on the shroomery at the beginning of the '08 summer. I spend a decent amount of time on this community looking at the Grow Mushrooms section, and at the Mushroom Cultivation, and Grow Logs section. After about 3 months, I had read up really well on everything, and started to help out some people who just started reading up. When I finally got home after summer, I ordered most of my things online, and got ready for a grow.


Preparation

I started out buying a few things online:


These were the main things that I got, everything else is not very important, and I wouldn't like to write everything down, since you probably won't read it, and you will notice it in my log anyways.

Here are the syringes, this was after inoculation.

The Strains are:
Brazilian
B+
F+
Golden Teacher
South Transkei
Orrisa India
The last two to the right are oyster strains:
Pink Oyster
Phoenix Oyster

Making a Glove Box

My glove box is super simple, and has proven to be very effective, anyone could use cardboard, but I chose to use plastic since it lasts for a lot longer.

I cut off two holes as big as my arm, and my friend helped my too, by heating up a metal stick, and cutting the tracing we did with a pen. Then we duct taped the sides of the hole, and took a strong plastic bag, and made it into sleeves which we taped to the holes.

The reason why I did this was so that it wouldn't allow contaminations floating around to get inside as easily, they could hit the outside of the sleeve, but would be hard to get inside. Also since I flame sterilize I still needed to get my hand out, so this allowed me to do this. The glove box could then be used as normal, by sticking in the sleeves and closing them, then using 91% isopropyl alcohol on the sides, and then spraying with lysol(or bleach spray), and let them work for about 10 minutes.


Substrate and Sterlization

Regular PF substrate was used; 2 cups vermiculite, then 1 cup water, then 1 cup brown rice flour. As RR says, it's important to put water to vermiculite before adding BRF because that way it can coat it better. I filled up the jars except for 1/2 and inch, where I put dry vermiculite, which serves as a contamination barrier. Then I put some tyvek, which serves as an even better contamination and is 99% contamination proof.

Inoculation(9/9/08)

For inoculation I used my GB of course, and this was done in a small bathroom. I sprayed the shit out of it with lysol, and let it cool for 15 minutes. The glove box was cleaned on the sides with isopropyl and then sprayed with lysol. The sleeves were closed with a rubber band. After that, time for inoculation. I flame sterilized every 4 jars, and wiped in between every jar. This took me about 2 hours to do every single one. After I was done they were placed into the box, and labeled appropiately:

The oyster mushrooms were placed in my basement in a cooler temperature around 74F.


My others cubensis jars were placed on top of my closet, at a temperature of about  77F. I wanted the temperature to be a little bit hotter, so today I'm going to turn my small bathroom into an incubation chamber, with a small heater.

Colonization - UPDATE(09/18/08)
9 days since inoculation

I know I said I would update Tuesday but there were some camera problems, and I'd rather not use a phone camera. But anyways, here are the updates. 8 of them got contaminated, surprisingly they were all the cakes belonging to two strains: South African Transkei, and F+. I inculcated both of those strains in the same GB run, so it had to do something with the overall cleanness of my GB before that inoculation. Anyways, less talk more rock, here are some pictures...

Now moving on to some oysters...I have no idea which ones are Phoenix and which ones are Pink, but I didn't really care because I thought it would be cool anyways when they're in the FC and there's a variety of different types of oysters, and different colors in the FC. :grin:

This is what I thought was a contamination, but later realized that the mycellium was growing just fine, and that slight discoloration didn't spread:

Anyways, that's the update for today, and I will update you guys as soon as the germinating jars start colonizing.

UPDATE(10/06/08)

Sooooo...
A little bit of an update, I know it's been a LONG time, but my jars weren't looking so good(and still aren't), but most of them are ready for birthing so let's see some pictures!

Family picture! I'm so pissed, I could've been eating these by now! Perlite is so hard to find in my area. :rolleyes:


Only 2/18 Oyster jars stalled, turned them upside down, and they're doing much better. To the right is another picture of some weird blotch thing, but doesn't look like it's growing or anything, so it's probably just a wet spot or something:


Some more jars I inoculated about a week ago maybe. B+(left), GT(right). I'm really angry, the reason why I had to re-do these jars was because I messed up 14 cakes!!! Used bleach instead of lysol, and I waited only  about 5 minutes(it worked fine with lysol).


This is one of the jars that survived the GB bleach run, very impressed! Most of the other got contaminated at all 4 inoculation points!


Some weird stuff on one of my GT jars, probably contaminated...this is why I prefer putting tyvek on top.


Finally! One of the cubensis jars that made the great stall! This one is Orrisa or Brazilian, at this point I was so angry because they stalled that I put the jars back in any random place and totally forgot which jars were which :mad2:. But oh well I'm not experience enough to do MS syringes. :smirk:

And finally! Sorry I haven't introduced you guys to my TiT! It keeps temperature at 81F, same temperature as Roadkill uses.


Well, that's it.
I would provide more info on how many jars I have left and blah, but I'm just too disapointed with myself at this point to look back at the cakes...

Fruiting 10/2/08

Well it was finally time to put them in the fruiting chamber, at first I tried simply using perlite, but then I took my ultrasonic and attached a garden hose to it, and voila, my hygrometer first read 99%, then 'wet'(100%), and then shut off...awesome.

Now there is some bad news, and good news, and on with the bad news first. ONLY 7 OUT OF 30 OF MY CUBE JARS MADE IT TO FRUITING. 22 MADE IT TO BIRTHING, BUT ALL EXCEPT 7 SMELLED LIKE ROTTEN APPLES. Tossed them out, even if they were 'good' I could not stand the smell, honestly one of the worst things I've smelled in my life.

The good news however is...OYSTER PINS!

This is my setup. The lamp is an aqarium hood with a 6,700k daylight flourescent bulb.


Incredibly simple to make, just duct taped it, took less than 30 seconds.

A little bit about my set up:

As I mentioned I use an ultrasonic humidifier and all I did was cut off part of a garden hose and duct tape it.
I use a 6,700k light with an aqarium hood, I got it at Petsmart and it was fairly expensive.
It has A LOT of holes, I use polyfill about 50/50 on the holes.
I fan 5-8 times a day.
I leave the light on a 12/12 light cycle starting at 6pm and ending at 6am.
There are 5 inches of perlite that I use anyways.

Any more information will gladly be provided message me or post here, I'll be glad to tell people about my grow and teks I used.

Expect another update before harvest for the oysters, or when pins show up on my cubes...hopefully this week.

Looks good to me but the bathroom is about the dirtiest room in the house.

Well, I never really use this bathroom for anything, and I couldn't spray any other part of the house down with lysol. It's better than a "clean" part of the house that isn't sprayed with lysol. The bathroom also had no air circulation.
Besides, I checked today on the jars, and all the one's that germinated have no visible contamination.

Wow Go big or go home eh! Looks like you invested a lot of start up. I started small with Oyster Mushroom logs (Precolonized cakes that all ya gotta do is birth and watch). I first learned how mushrooms grow and how to maintain thing like humidity and temp. I've since moved on. I just hope everything works well. It sounds like you're doing your research and starting right. Contams didn't start showing up for me after about 10 days (BRF/Verm PF tek). Good Luck. keep us posted.

Oh, I did have one complaint but it may be nothing. The picture of the syringes, you laid em all out side by side with no identifying characteristics are you sure that Syringe 1 is Syringe 1 and not 2-7?

I actually do have them labeled, but it's not on the side where the ccl mark is, because then I wouldn't be able to see how much I injected now would I. :wink:
Take a look at the last(oyster) syringe to the right, it says Phoenix Oyster, a lot of the ink got rubbed off by the alcohol, but it's still visible.

I guess I should get on to some better form of labeling, maybe labeled paper under tape.

Psilocybe comes from the Greek root "psilos" meaning bald head and Cubensis because it was first recognized as a new species from specimens collected in Cuba.

Nice work, flame your needle before inoculating each substrate to prevent cross contamination.

Your sterile procedures are a bit over the top IMO, i never spent that much time cleaning the room i was inoculating in when using a glovebox. I dont even bleach/alcohol my glovebox, just clean it with soapy water transfer all my materials in, leave for 10 minutes and then carry out my procedures.

Still air boxes work because theres little/no air movement to kick up contaminants into the air, in other words they stay settles on the bottom.

I have a few tubs, I'm definately going for a monotub hole type setup, with polyfill and everything, except I'm either using perlite, or connecting one of my humidifiers.

For the oysters, a normal shotgun FC setup will do the job...
Maybe a little bit more holes, because I'm really busy and don't have so much time to fan and mist.

This seems like a ringing endorsement for flame sterilizing between every jar. I don't think it's a coincidence that you have four contaminated jars of each... I can see why you cut corners, though with all those jars to noc up. I agree with the poster that said the mad cleaning and spraying is unnecessary, though. At least it hasn't been necessary in my case.

Anyway, best of luck on your most ambitious first project!

I actually figured out what was the problem.

During the last GB run, I ran out of lysol and I used bleach. I always wait only 5 minutes with lysol, and all seems good, but when I waited 5 minutes with bleach they got contaminated.

I even did a little experiment to see what was my flaw in sterile procedures, and I did the same thing in two GB runs except I replaced lysol with bleach, and only one made it. As opposed to lysol where they all made it 100% contaminant free.

I did think about using bags actually...
...after I bought the jars.

Probably going to get them soon anyways, since santa might bring me an AA this christmas. :tongue:

Now there is some bad news, and good news, and on with the bad news first. ONLY 7 OUT OF 30 OF MY CUBE JARS MADE IT TO FRUITING. 22 MADE IT TO BIRTHING, BUT ALL EXCEPT 7 SMELLED LIKE ROTTEN APPLES. Tossed them out, even if they were 'good' I could not stand the smell, honestly one of the worst things I've smelled in my life.

The good news however is...OYSTER PINS!

This is my setup. The lamp is an aqarium hood with a 6,700k daylight flourescent bulb.


Incredibly simple to make, just duct taped it, took less than 30 seconds.

Surprise! Mad pins! This one is an phoenix oyster.

The most bottom ones on the left picture are the cubensis, which have not started pining, but with patience they hopefuly will.

This is very interesting actually, I think I somehow mixed pink and phoenix oyster in one jar...looks good though.

A little bit about my set up:

As I mentioned I use an ultrasonic humidifier and all I did was cut off part of a garden hose and duct tape it.
I use a 6,700k light with an aqarium hood, I got it at Petsmart and it was fairly expensive.
It has A LOT of holes, I use polyfill about 50/50 on the holes.
I fan 5-8 times a day.
I leave the light on a 12/12 light cycle starting at 6pm and ending at 6am.
There are 5 inches of perlite that I use anyways.

Any more information will gladly be provided message me or post here, I'll be glad to tell people about my grow and teks I used.

Expect another update before harvest for the oysters, or when pins show up on my cubes...hopefully this week.
