First Petri Dish Attempt

Well here are some pic of my first attempt at agar.. I knocked them up and checked them today, and I see like 7 or 8 little spots of white.. When looking at them close up they are not really rope like they are like a little spot of cotton, they are fluffy.. So what do you think, go or not good ??

i can't stop looking at your avatar, and wondering what she is saying.... you have a full size version perhaps?
as for the spots, my first ones currently look just like yours here. I think it is mold. I think mine are mold...
did you inoculate with spore syringe?
next time I'm gonna take a single colonized grain and inoculate with that... my solution just seemed like it ran all over the agar, which I suspect yours did too.
a single colonized grain should make isolating rhizo easier too I beleive... and it won't run all over the place, it will stay in the center.
Lets wait for some knowledgable folks to answer tho!

in my mind she is saying dirty dirty things - so yes I CARE! lol
don't listen to my advice, if these folks say good its good. Upon checking mine again my second generation looks more rhizo... so I think the cotton is just 100s or thousands of substrains right on top of each other, giving the appearance of cotton...
question here, don't mean to jack but this is useful...
How do you know when it is mold? I've heard agar can get mold that is white and cottony. Does it just take time to tell mold vs mycelium?
I just got the RR video today in the friggin mail!!! I got it almost exclusively for the agar section...

lol the only time I look at her mouth is when my dick is in it.. I am not isolating a strain per say, well not yet.. The is just some myco scrapped off of a corn kernel from a g2g jar I had.. But when I get something I decide I like I will isolate it.. Just playing around with agar to try and get the hand of it..
I don't have a blown up picture of the girl but I will try and find one..

I always thought she was saying "Put it on Bitch" but thats just me probably. I like to think I can somewhat accurately read lips, but sometimes that just becomes Me eavesdropping incorrectly
Those trays look good, but you'll want to begin isolating away from any contams as soon as you see growth. Isolate from the farthest away side from the contams, and pick the most aggressive rhizomes

I'm going to switch to the powder and make mine in a long neck whickey like bottle to be easier to pour...
how much do you think the pound of mae will make? its like less than $40 from several of our vendors. Seems to me this is a shit ton of agar is it not?

I think you mean MEA.
Edit: I thought you bought premixed MEA and were going to buy 1 pound of agar agar powder to make your own agar mix later. 1 pound of premixed MEA and one pound of agar agar are 2 totally different things. My bad. I should have read instead of skimming through.

the pound will make... 40g to 1 L water, so... over 12 L of media and a L of media will fill some 50 dishes. so 50x12=600 dishes.
and IME using more like 30g per L of water works better. I have had too many times where I PC the premade MEA with 40g pre L, and it never sets up due to over heating I think... not sure tho

A pound of MEA powder should make over 11 liters of media and a liter easily makes 50 plates. So you could make over 550 plates with that.
It's 40g per Liter BTW.
You could just buy agar agar powder and malt extract from a brewers supply. You could also get dextrose from the brewers supply and make PDA. It's good to switch up agar recipes when storing a culture long term. I believe it's common practice to switch up the media used every 6 months.
