RR's Tyvek Sleeve Tek and Mycobag Size..... Cooling Time...

I've received a bunch of small sized bags from Sporeworks and now I am wondering if I will be able to use them with the Tyvek Sleeves. I don't have any Tyvek sleeves on hand right now, but I've used them before. I was planning to order the sleeves, but now it seems that it may be a tight fit (from memory). I don't want to order a bunch that I can't use.
I thought the Tyvek sleeve tek would work well. I can get access to a university autoclave on Monday. And then let them cool till Wednesday when I can inoculate them in front of a hood.
It would be nice to possibly sterilize and inoculate on the same day.... But I would have to do the inoculation work just four hours after the media went into the autoclave.
Would this be enough time for bags, grain jars, or even liquid cultures to cool before inoculation?
And if I get my shit together on a Wednesday would the following Monday simply be too long to wait for inoculation?

If you can cool them down in front of the hood fan they should cool down quickly. The grain jars should cool down in that time, I don't know about the other stuff.
And if I get my shit together on a Wednesday would the following Monday simply be too long to wait for inoculation?
I don't think so, at least not for the grain jars, I keep sterilized jars of grain in plastic bags for weeks.

You can always fold the tyvek sleeves if necessary. As the vacuum tries to form in the bags, they'll pull tight around the tyvek, thus remaining sealed. Another trick if you have tyvek coveralls, is to zip the bags up inside the coveralls as they cool. I've even placed them inside the coveralls in the PC prior to sterilization, so that they're already contained as the PC cools.
As falcon said, if you cool them in front of a flowhood, it's not necessary, but that becomes difficult if you're doing several batches in the PC and plan to inoculate them all at once a few days later. RR

Thanks both. The whole "folding" concept came to my tired mind about an hour after posting. Embarassing.
I'm going to show up with some quart jars of grain Wednesday morning then- hopefully get them sterilized and then inoculated before I leave the lab. Then, I'll have master spawn to spread around for my experiment (whatever that is) in bags later.
Geez, I just washed up my grains really well and added the gypsum to the soak. It turned the water really cloudy. The package said 94% gypsum. I wonder what that other 6% was?

Was it the pelleted stuff used to loosen soil? I get that too. Maybe it's clay or something. I boil my grains in the gypsum water as per RR's video instructions and I haven't had many clumping or busted grains, at least when I dry the grains while they're still hot and spread them out over a flat surface. It's not easy to dry them in a colander using residual heat, and leaving them to drain on their own tends to leave a lot of moisture, too.
Must be nice to have access to lab equipment!
