sterilizing MEA in the microwave?

is this possible? any suggestions? could i try it in a plastic autoclavable flak? i would plan to add peroxide also

yes it will work, i use jam jars. i put in the amount i need already melted and mixed agar. then i mark the level on the jar and then i add in aanother 100ml of water. i put the cap on loosely and nuke for about 3 mins on full. then about 20mins on defrost just to keep it simmering. by then the water will have reduced to the line i marked. if it is not at the line just nuke some more. if past the line add more water and start all over again!

execllent, thank you blackout! i will try it today. i assume you use a nonmetal lid? do you think i could use the plastic (autoclavable) flask, or is there a risk it could melt?

no i used a metal lid and always have done and have never had a problem with it (cue the flames!).
microwaves are reflected by metal objects in a microwave. small metal objects like pins, cds, tinfoil store the energy and release it in the form of sparks or arcs. if you have a large metal object (like a lid) and a substance that will absorb the microwaves (the agar) you wont have a problem. lots of microwaves have a turntable made of metal and have no problems. i did an indepth course on dielectic heating (microwaves) in university and they dispelled many myths on the first day.
the flask should be fine although high sugar mixes get well above 100C when boiling i dont think a 4% sugar solution should be a problem

If you read about microwave cookery in your manual, you will see information about covering large pieces of meat strategically with pieces of foil to distribute heat, but I've never personally attempted using metal in the microwave.... not that this is even really directly related to this post ;p

if making peroxidated agar you can just make it in a small saucepan on the hob.
just mix the ingredients except the h2o2 into cold water, bring to the boil for a minute or less, allow to cool to ~125F and add the h2o2, stir well et voila!

at last somebody bothered to read!! they probably recommended folding it over into a thicker piece and not having sharp edges hanging out. every microwaving metal hater take note! or just ignore as you probably will...

thanks for the ideas. i poured some petris yesterday using the microwave + added peroxide. champ, i take note of your point, and will pour some peroxidated MEA w/o nuking. now, for spore germination, i remember seeing you had a tek for doing that, i.e. with the cardboard. tips for this? any cardboard? moisture content? the side of the cboard with spores is then placed on the agar. on which side? spores up or down (so they are in contact with both the agar and the cboard??

no, not corrugated cardbord. thick cardboard, like the stuff that comes on the back of notebooks etc. I believe the technical term is "pulpboard";)

soak for 10 minutes in hot water, or if you're germinating non-woodlovers, honeywater or the like

spore side up. the point of the cardboard is to protect the ungerminated spores from the peroxide in the agar, while providing the smallest possible surface area for contaminants to fall on. also, if the cardboard is soaked in plain water it is pretty much contam resistant.

The scary thing about microwave is ............... some ........... not all have a tendency to heat from the middle out causing some areas to get hotter than others , the problem with that is ........ if you go over 250*F the agar can become carmelized thus ruining it . PC or AC is the way to go , that or do the boil method .

EXCELLENT! thank you champ, i'm getting right on with it, now i have all the info i was looking for. i'll keep you posted as to results. i can tell you now that i don't seem to be experiencing any caramelization problem. i've been following tblackout's suggestions above (3 mins on high, then 20 ms on defrost to reduce excess water) and everything seems ok.

actually, when you say "the smallest surface" etc, how small? you mean the cboard would not be a disk with a circumference close to that of the petri, but a much, much smaller circumf? how small?

Why are you putting whole spore prints on agar ? I only ask cause the , method I used was smearing spores on agar surface with an innoculating loop . I have more spores left over than I know what to do with . :cool: 

Sorry , just realized this is a tech I have not seen , and ASS U ME D it was similar to method I performed .

the size of disk that a hole punch for putting paper into ring binders makes, ie ~5mm, though smaller would conceivably be better, up to a point....
the disc is just there to provide a peroxide free patch for the spores to germinate on, and once they're geminated, and thus able to break down peroxide, the mycelium can "step off" the disc onto the agar and colonise it.

ok, did a couple of experiments today as follows:

1.MEA/nuked +peroxide, used to pour petris
a. inoc with psy cyan myc water; and
b. inoc with psy cyan spores from print over a cardboard disk. the disk was soaked in slightly nutrient solution, and nuked for 3ms; and
c. same as b., above but with psy cub/huautla print; and
2.MEA/nuked W/O peroxide used to pour petris
a. inoc w psy cyan spores/print; and
b. inoc w psy cub huautla spores/print.

i guess that's it for today. time to go watch startrek

no, soaked in a jar of slightly nutrient solution and nuked at the same time. after nuking, i simply discarded the excess water. it looked ok
