A little fun with anastomosis

A little while back there was discussion on mixing to cube strains and "make a new strain". Just wanted to post one of my cakes that I used two strains on. The strains were Large Fruit and PF redspore. Here is some pics. I can see the difference between the species, its really cool! Thanks RR for the info and inspiration!

That is very cool - can see the similarities to each of the "parents" but it's definitely its own being. Now to see if it holds the traits of each you want - looks great

Just made up a syringe with half and half, it was pretty easy. But its fun as shit! Yea I am gonna try and keep this one going, I love the way it looks! It looks like a Transkei but with a red print! Even the veil opened up like Transkei does, its crazy!

For those of you who are asking, sometimes two strains will mix, sometimes they won't. However, they NEVER fight as was commonly thought a few years ago. If spores were used, the monokaryons may have crossed, or it's possible that later on the dikaryons crossed by anastomosis, or possibly even a combination of both. Nice sporeprint. RR

Could you do the same thing with mixed myc? Or does this only work with spores?

Interesting...
If it's even possible, what would it take to pull a single spore from each of two strains and get them together in a petri? A good microscope and some sort of "fine-point" inoculating loop??? And a very steady hand (or robotic arm???) Or is it get as few of each as you can and cross fingers?
I'd like to play with this idea.

two different strains*and use new clean foil that had a quick wipe down with rubbing alcohol. its a good print but it looks like you picked up some left over wrapping from bk.

Trying to cross two individual spores rarely works. You're better off using the bacteriological swipe method or massive spore dilution to get single spores and then transfer monokaryotic mycelium away to other dishes to work with. For massive spore dilution, simply squirt a few drops from a syringe into a large glass of sterile water, and then draw the diluted solution into a fresh, clean syringe. However, such can easily lead to non-fruiting strains.
Another method is to make a spore print, and then lay a cap from another strain over the first print and let it print on top of it. Then, make a syringe as normal.
To breed dikaryons, place two isolated strains on agar an inch or so apart and let them grow together. If a third sector opens up at the point where they meet, it is a cross of the two strains via anastomosis. RR
