    #902638 - 09/23/02 07:38 PM (16 years, 2 months ago)

is it possible to clone from a mushy to agar? i've done multispore and mycilium transfer, but never cloned to one. just curious. i want the characteristics of this particular mushy, but want to see if i can isolate it further to maybe enhanse it a little more. thanks

yea sure...

Just take the fresh mushrooms, cut the stem into a cylinder, use a razor to peel back layers around the stem like a banana, to get the fresh sterile (semisterile) inner stem portion, and toss it on the agar.

In fact you may not even have to select the best mycelium (like when you did multi) , that is advantage to doing clones , but it is reccomended that you return to spore/agar if any signs of degredation shows up . That is if strong rhizmorphic growth becomes hard to obtain from culture .
Good luck

hi yes it is, pick your fastest biggest sample if you don't have a flowhood you can use hydrogen peroxide in your agar mix to help with contamination. But you still need to be sterile as possible.

If you add H2o2 to the agar mix, it will revert back to water during the sterilization run.
