I'm not sure how many people in this forum also view The Pub, so I figured I'd let people here know about my latest microscopy adventures.
Rather than repost the pictures here, I'll provide you with a link to where I've already posted them. I'm considering just having the post moved here... Pub folks don't seem to interested in microscopy.

If anyone here can ID anything in my latest post, it'd be appreciated. I hate looking at stuff and not knowing what it is. I really don't even know where to begin looking up these organisms.

the other day i was stoned and bored so i decided to see what my sperm looks like. it was pretty interesting.

I have a question for the people who use microscopes.
I've never used a microscope that has a movable light source and an aperture... Mine does. I've tried to figure it out using my knowledge of how photography works. I don't think I have it quite right yet.
From what I've found, low magnification should use the light source far away and the aperture should be open all the way. High magnification uses a very tight aperture and the light source is right up against the slide.
Does that sound about right? Do you have any pointers for me?

Hmmm, the microscopes I mostly use are automatic but even then I haven't noticed the light source moving. On the manual ones I don't have to move the light. High power objectives are up close to the specimen and lower power ones are further away, but the light source is fixed in the base of the scope. The light passes through a condenser just underneath the stage before it hits the specimen, and usually the condenser has a slider or two that changes things. I usually fiddle around with that until it gives an image that I like.
Actually there's also a condenser focusing knob, perhaps that's what you're thinking about? That sets up the Kohler illumination - I'm not sure if all scopes have this, perhaps they do but I never touch it on the manual ones. To set up the Kohler, you have to shrink the field stop (aka field diaphragm, this may be the aperture you referred to, because that's what it looks like) until it's visible, then use the condenser focus to make the field stop appear sharp. Then expand the field stop back out of visible range again. Like this:
 Doing that gives you much better images. But as I said, I don't know if all scopes use Kohler.

Ah ha! That's exactly what it is. Thanks zouden. It looks a lot more crisp now.
I'm guessing you only have to set up the Kohler at a low magnification because that's the only time you can see it, correct?

I'm not sure - on the scopes I have at work I can do it on any magnification, but I think once it's set up for one it works for all of the objectives. To be honest I often forget to do it unless it's obviously wrong. It's not as important for fluorescent microscopy which is what I mostly do.

I think the Kohler is most effective at high magnifications like 1000x, its necessary to have it set up right to get true 1000x magnification.
For high magnification the condenser should be really high up but not so high that it touches the bottom of the slide and scratches the lens. For the best resolution people put a drop of oil between the condenser and the bottom of the slide, and another between the cover slip and the oil objective. I usually skip the lower oil so there is less to clean and it doesn't seem to help much anyway, but maybe on some scopes it would.
I sold my microscope to cactu for 2000 pesos and I ordered a new one that has phase contrast and darkfield, I have been checking my po box every day for that.
