Planning A Class Project. A few questions.

Hey. Im trying to come up with some kind of experiment for my biology class and I would like it to involve mycology.
Nothing major. Probably, I will grow some kind of oyster mushroom and manipulate the media in some way, and compare growth.
I figure the results will be more meaningful if I can use a single isolate. So, my first question is what is the best shortcut to getting to a single isolate? If I buy a syringe or a wedge will that likely be an isolate?
Secondly, I am wondering how easy it is to control fruiting on The King Tuber Oyster, so that I would only have sclerotium to quantify instead of mushrooms?

*Spores Syringe- Multi genes. *Culture wedge- Depends if it is from clone or from spores. - Wedge from sprores- Multi genes. - Wedge from clone- 1 type of genes.
Ofcourse a clone from good healthy mushrooms is preferable

You can get fresh king oyster at Asian markets. It is a big mushroom and very easy to clone. Make jars of sterile grain and throw bits of inner tissue from the same mushroom into them, make several from the same one and be prepared to loose some to contams. Don't change the food, the most dramatic effect can be seen by altering the fruiting conditions, specifically the addition of CO2 or by limiting fresh air exchanges which will keep CO2 in. Skinny tall shrooms and short stumpy shrooms will be the two results, much more dramatic than more or less sclerotium due to the addition of some mineral or whatever.

Buy one, or clone the mushroom that you would like to isolate.
