Life span of mycelium in petri dish & mycelium to jar inoculation???

Hey,
If I am using MEA petri dishes to culture mycelium, spawned from a syringe, how long do I have to use them once they are fully colonized before they go bad or rot or whatever?
Can they be stored in the fridge or something to slow their decay (if they do decay, that is)?
I want to inoculate some pint jars but don't want to buy a whole s-load of syringes, so I want to use the mycelium to inoculate the jars. You can do that, right? I have a GB and pressure cooker so I am not too worried about contaminates. Anybody have exp in that method?
Also, how long does it take on average for a petri to be fully colonized?
If you can't tell I am new at this so any over-explanations will be welcome.

Petris usually take a week or 2 to colonize completely once you see myc start growing in my experience, but it can vary. Some mycelium will grow very rapidly and rhizomorphic (like little ropes) and some mycelium can be cottony and slow growing. Typically, you want to transfer the ropey growth to more dishes until you get one "strain" that doesn't have different sectors.
If I'm not doing any more transfers, I usually put my dishes in the fridge once they are about 3 quarters colonized. I have used 2+ month old mycelium from dishes successfully, but for really long term storage I would try to transfer some of the myc into sterile distilled water and seal it in a vial like a vacutainer in the fridge. Roger Rabbit also mentions something about using popsicle sticks in a vial in the same manner.
Yes, you can definitely use wedges from the petri dishes to inoculate your jars in a glove box. You can also transfer the wedges to a liquid culture in the same manner to really multiply your supply of inoculant.
I would highly recommend using the search feature and searching through the 'advanced' cultivation forum for lots of information on doing agar work. Roger Rabbit and Workman are 2 members that come to mind that have posted some really good info on the topic in that forum.
Man, that felt like writing a novel! Anyway, good luck with your project. It's good that you're going "old school" with agar instead of just injecting spores into brf/verm cakes

Actually I am planning to start with brf/verm cakes, just because I'm new to this and want the least chance of contamination, however I hope to switch to the organic rye method once I get a hang of it. I just don't want to use syringes because I feel that with the mycelium I will have more control over the entire process, and because they are so much more expensive. What is the best method you recommend for the bulk method (rye under a layer of vermiculite or whatever)?
Also does it matter what size vermiculite I use, coarse or fine, or does that not matter at all? Also does anyone know of what trade names vermiculite is sold under if any?

Fine verm works best with the PF tek IMO? Some people like coarse verm better so you will have to experiment to find which one you like best. That will go with just about everything you do in mycology. There are somethings that you should do just as the teks say.
But being new you should stick with the PF tek and do it to a T.
