Pressure Cooking Agar
    #876661 - 09/11/02 04:55 AM (21 years, 7 months ago)

I'm about ready to PC some agar for my first time. I have a 500 ml erlymyer flask to cook it up in. I read to plug the top of the flask with non absorbant cotton and then cover with aluminum foil. I was wondering if cotton balls were sutible then alum foil. Or is there some kind of other cotton I need to use. Could I use polyfill then alum foli? Or can I just use aluminum foil. What do you all use? Thanks.

i use aluminum foil alone
but a also use a PCer that forms a vacuum what i think is the use of the cotton /polyfill/wtf ever is to slow the rush of contaminated air into the flask on cooling

but i dont think that there a huge difference in types of cotton

Be sure to let the pressure return to normal slowly or you will have an almost empty flask and a big mess to clean.

I just use a Quart jar with a filter disc and a lid with a hole in it.

Just got back from Meijers. To my suprise I found rolled cotton, just what I was looking for, right next to the cotton balls. So I'll use that and aluminum foil. Thanks for the replies guys.

it's best to use hygrophobic cotton or the cotton may be all wet and soaked. Cap with tinfoil.

Very important note, DO NOT fill the 500 ml flask with more than 300 ml of agar or else it will boil out the flask. This will give a huge mess
