Re: Question on agar culture

Either the culture should be completely sealed, or it should be in some sort of sterile humid enviornment; otherwise it will dry out and get contaminated. Spores should germinate within a few days.

It?s a good idea to seal your agar containers with a cut piece of saran wrap like on picture. This way gases can exchange but no contaminants.

I've tried starting an agar culture with spores that I know are viable, after a week..still nothing. My jars were sealed up air tight.Maybe I should try the saran wrap thing or get some petri dishes instead of my pyrex bowls.
I guess this proves that mycelium needs at least some air circulation, unless theres' some other variable I'm not taking into consideration.
Anyone else have any other thoughts on this?

Yes, you must not seal the petris compleately, they need some gas exchange. Seal them with saran wrap.

So, I take the seal of the Saran Wrap is just imperfect enough to not be airtight? How did you cut the whole roll of Saran Wrap?

When you seal the petry dish, strech the saran a little, it will then take the form of the petri and seal it very tight.
Polyethilene is tight against water and small particles but leaves gases through.

I cut the piece off with a sharp knive. Watch your fingers!

Oh, it does, it does. Not much, but it does.

The whole side of the petridish is supposed to be sealed, so the whole thing is dust tight but not gas tight.

I found that I might clear some things up.
The plastic foil I use to seal petri dishes IS NOT the original Saran Wrap tm. it is a thin plastic foil made of PE(poly ethylene).
Poly ethylene leaves gases through.

Saran Wrap on the other side is made from PVDC(polyvinylidene chloride).
The source says that ...The result is a barrier against oxygen, moisture, chemicals and heat-qualities used to protect food, consumer and industrial products.....

So it might realy be that the original Saran Wrap won?t work.

Try finding some foil made of PE.

More info on Saran Wrap.

I bought a roll of Para film (2 inchs large) from Fungi.com-(elastic wax like film). It works well. I can't remember the price, it should be $25-30 for 250ft. About Gas exchange in petri for Mycelium growing is not sure to need. I recommend wrap it tight with film anyway, if U keep them in unstelirized condition.

I've heard good things about using electricians tape as a dish wrapping. Just a thought.

i use the baby bottle for agar stabs.cut the nipple tip off and stuff with aquarium filter floss or cotton.this allows for ventilation and maintains sterility.plus spores can be injected through nipple.
