big inoculation problem

so i got all my jars in my glove box...go to inoculate...and my inoculation holes are not big enof!!! wtf... how do i go about fixing this problem...i cant resteam these jars, im not off by a hole lot but ive been trying to be very contam free and now what?! take a hammer and nail back to them?

i've had this happen before, better be very careful and bore the holes out a little bit more with a larger nail. =/

The time I had it happen I had no problems, all colonized fine, but it still sucks.

should i take the lid off?! or hammer down into...im using PF tek so i have verm layer on top

If it were me I would get a bigger nail and flame (and/or) wipe it down with alcohol in between jars.


and yeah definitely leave the lids on there

Don't use a hammer just twist the nail and work it throgh the hole.

too late, i lightly tapped the hammer to make the holes bigger. it worked fine i suppose and i flamed/wiped down nail with alcohol. strange thing my first time inoculating and when i flamed the needle with candle...it started to like shoot out the end of the syringe. i think it could have been air, but it was hard to read how much i was inoculating since the syringe had a big air bubble in it, but i shook it well and inoculated all 4 areas. I lyosol'd the fk out of the area about 3 times b4 i inoculated, and when i tried to fit my syringe in glove box it was too small, so i put the jars down inside the lid but left it open and did my work that way. im hoping i don't get contams, if i do i will know next time to be better prepared with my glove box. but other than that i was sterile. :smile:

I DO NOT RECOMMEND THIS IN ANYWAY!!!! I don't even use a glove box to noc up my jars I flame before the first hole of the jar and wipe with 70% iso between each hole and i never get contams in my pf jars. You should be fine. I would not use a candle to flame. You can make an alcohol lamp very easily with a small jar with cotton wick that you make with cotton balls.

I had this happen but my needle was pretty stout so i just used a little extra force and shoved it in the hole
