another spore solution que?
    #8625794 - 07/12/08 04:56 PM (12 years, 10 months ago)

i will be doing 5 halfgallon jars 3/4 full with wbs, now, with 10ml of solution how should this be divided up for a fast colonize time? also will i have any left after these? and i know to much will hurt them, but what would to little do besides slow the growth? my first thought was 2ml per jar. thanks to all that will help with this.

That's alot of WBS for 10ml of spore solution. 2cc per jar is pushing it.

so more? like how much? or should i lower the load?

more importantly, what would be ideal for one of the jars 3/4 full?

I use around 2 ml per pint jar, and may be overdoing it, but I would be concerned about using so little for a half gallon jar.

My temptation would be to inoculate your 3/4 full half gallon jar with at least 3 or 4 ml of spore solution.

Anybody else?

inoculate each jar in four different but equally spaced spots, 1/2 ml in each spot for each jar.

he's not doing pftek he's doing grains, and you should be fine with 3-4ml per jar, i'd lighten the load and only do like 3 jars with 3.25ml each.

make an LC in the future even from multispore to make your syringes go further.

After you noc those big girls up you need to shake the shit out of them to make sure you get that juice all over or the will take for ever to colonize. There already going to take a long time because of the size. In my experience. Happy growing and keep us posted.

Shaking them immediately after innoculating with multispore can actually cause them to take longer to colonize.

Spores need to germinate, meet up with another germinated spore, and transfer genetic material to become dikaryotic mycelium, which is the rhizomorphic type that produces fruits.

If you shake the crap out of them and spread the spores out it will take longer for the monokaryotic myc to reach the dikaryotic stage.

this is absolutely correct. Which is why you want 1ml of spore solution ran down one side, and left alone until it ropes up at least 30% of the grain, then shake. If not you get monokaryotic tomentose mycelium.

monokaryotic myc is tomentose and produces very poor pinsets, best i've seen monokaryotic myc fruit has been small clusters sporadically at best, deinitely affecting yield beyond 70% or greater, but, it will fruit.
You can tell this type of mycelium easily, there is an absence of rhizomorphs obviously, and it's fluffy and cottony like snowballs or something.

I stand corrected. I just use LC's these days and forget people use ms. You know one of those "If I use LC doesn't everybody?" things.

ive personally always had great luck with shaking my grain jars after inoculation.  i just recentley (after some reading) decided to try not shaking the jars after inoculation.  this is what my jars look like exaclty 1 week after inoculation and no shaking.  pretty weak. see how its all concentrated at the bottom.  and this was with 3 cc's of solution

now i dont have any pics atm of jars after 1 week when i did shake after inoculation but i will be innoculating some more grain tomorrow and shaking those and i planned on posting a thread just to show that i personally get faster colonization times when i shake after inoculation.  in as long as ive been doing grians ive always shook after inoculation and i know for a fact that i have more grotwth after 1 week than the pics posted above where i did not shake.  just thought id throw this out there.

you first need to let your MYC stabilize, then shake. I let mine grow until I can visualize clumps, then I shake, then wait untill almost 100%, then shake/break if you still have uncolonized patches. It's all about patience.

In the past I have shaken jars immediately after inoculation and than switched to not shaking them, never noticed all that much of a difference with colonization time to be honest but it makes perfect sense that not shaking would be better so if I were to make jars now that is what I would do
