Can anybody tell me what this is?(PIC)

A couple of my petris have some stains on the agar, im sure that it isnt contamination because when it coagulated it had it already.....and it didnt grow.

here is a picture of it :

Hope somebody can give me some feedback

If you're using homemade agar, it could easily be undissolved solids in the mix. That's what it looks like to me.

I don't think so. Others may have had different experiences though.

This is what I do. After boiling and stirring the agar mixture, I strain the liquid into a canning jar. I then PC the jar to sterilize the agar for storage.

It looks very similar to bacterial contamination. I saw this before in failed attempts to clone wild shrooms and tossed it.

no it cant be a contamination because it was there as soon as the agar coagulated.

Yes, superficially it does appear to be a bacterial contaminant. What indicated to me that it wasn't was the fact that the "bacteria" definately appears to be under the surface of the agar and the fact that it was present immediately after the agar solidified. Whenever my agar projects are contaminated with bacteria (usually due to dirty tissue), the contamination is visible as a slightly raised layer on the surface of the agar. Since the bacteria produces a liquid, I'm assuming the protrusion is caused by surface tension.

Definitely looks either like bacterial contamination or yeast.
Watch it spread over the next days.

And it didnt get bigger or changed shape.

But ill just have to wait and see :wink:

why dont you just throw away those ones and use the other ones that dont have it... looks like bacterial to me as well... just throw it away no point in keeping it if you have others...

There is no way bacteria or yeast could appear within ten minutes of pouring liquid agar. I still think it's undissolved ingredients in the mix.

Be even more carefull when preparing agar.

How long did you sterilize the agar?

I did know it was a bad thing when culitivating shrooms :tongue:

But i thought maby because it was a little bit of the plate that is coverd with yiest that the other side may still be good to germinate the spores. and if the spores germinated, that i could transfer the wedge to another plate.

I sterilized for 30 min.

And i didnt use to much yiest (exactly 2 grams).

Dang, that really looks like bacteria. Motile bacteria. If it's as you say, it certainly couldn't have grown that quickly.

Can you give it a smell test? Even yeast should be obvious, bacteria should be downright foul smelling by now.

If some of the agar ingredients have fallen out of solution, you might want to double check the recipe and give it some extra swirling next time. That would be the first time I've ever seen a clean plate that looked that way.

Well 5 days has past and spores are germinated (There is a update about it in the growlog here : Mazatapec Growlog (inocul. agar.)(PIC'S) )

And im for sure now it isnt contamination, i think it is undisolved yeast.
Im for sure it isnt contamination,beacause:
1. it was there when it coagulated.
2. it doesnt get bigger.

I think my problem was that i didnt mix it that well.
i just shaked it a few times in a erlenmeyer flask.

So what did i learn from this,

Mix all the ingredients verry good :smile:

you poured your agar too hot water condensed on the top of the petri and driped back

its happened to me

made contamination pop up so i discarded
