Procedures:
All of these were from several month old sterile spore syringes.

Substrate is wild birdseed that has been highly successful in other cases. WBS was soaked for at least 12 hours, and no more than 24. Sterilization consisted of PCing at 15PSI for 1 hour.

Spawn jars are 1pt wide mouth jars, one 1qt regular mouth jar is included. Filter is one layer of Tyvek disposable coverall material.

Inoculation was done in a glove box sprayed with bleach/water solution; jars and equipment were wiped with 93% rubbing alcohol. Inoculation dates are either 5-28 or 6-30. Needle was flame sterilized before the first inoculation.

Casing material is verm and peat, roughly 40/60 by eye, respectively. Casings were loosely compacted and covered with foil, in which air holes were punched every 2-3 inches.

The Zoo

Specimen 1: When I removed the tin, the casing exuded an ammonia like smell and a slight earthy mushroom smell. Very little to no mycelium has reached the surface.


Specimen 2: The bottom of this casing is quite visibly wet, and smells like one would expect soggy wbs to smell and earthy mushroom scent. Trich. is prevalent and highly visible from the sides of this casing. Interestingly, this casing was quite warm to the touch, easily 10 deg F above room temperature.


Specimen 3: Noticeably soggy bottom, with an almost gelatinous look. Normal smell. A redish (possibly pink) contamination is visible from a different angle.


Specimens 4 through 8: These are from the same batch as the above, but were not cased. All have excess water collected at the bottom of the jar.

Specimen 4: A wet-spot like effect on the bottom and a black looking mold on top. The bottom was very dark brown to black as well.

Specimen 5: Wet-spot like effect on the bottom, the mycelium appears to be fighting it's way through.

Specimen 6: A surprisingly black liquid visible in the bottom half of this jar.

Specimen 7: This jar initially showed signs of Trich contamination, but has largely been taken over by the Mycelium. Some is still visible through the layers of mycelium. Is "any" too much trich?

Specimen 8: Heavily contaminated with Trich.

Observations and questions

Mushroom mycelium appears unable to colonize areas that are submerged in excess water. This water was not visible immediately after sterilization or necessarily before inoculation.

All areas with extra water took on a rusty color, which may be contamination or the coloration of the seed. Breaking up one of these jars shows that this rusty discoloration has a jelly/gelatinous structure and the mycelium growth looks otherwise healthy and strong even just below the surface of this rusty jelly. I suspect this is simply solidified starch colored by the WBS, the jars are not contaminated, and are safe to case. Thoughts?

Some of the jars have a distinctly black water in them, perhaps 4 of approximately 24 jars.

Every single one of my casings from this batch is contaminated, thoughts as to why?

I'm not convinced on the above.
# 1 seems to have suffered death by High temps and "maybe" the start of black mold
# 2 just seems to developing a common "gel like" metabolites and "bruising".  It look like it has been ready for fruiting for a while now.
# 3 could possibly be bacteria, but could just as easily be matabolites.
# 5 and 7 are begging to be spawned!
# 6 is screwed by mold.

The bottom line seems to be that there were high temps for long periods of time and you just didn't induce fruiting when you should have.  The only one that could possibly be the syringes fault is # 7.  All the others wouldn't have come close to where they're at if it was a contamed syringe.

Something I forgot to point out, is that #3 has frosted looking mycelium on top of the casing layer. I can't take a good enough picture to share, but it looks similar to frosted wheat.

Other than that, none of them have sent much, if any, mycelium to the surface.

I suspect part of my problem was having the grains too wet when I started. I also did not sterilize my casing material. I'll be airing my grains to dry a bit after simmering and sterilizing my casing on this next batch.
