Good pics of panaeolus mycelium
    #8533695 - 06/17/08 01:32 PM (15 years, 10 months ago)

I am having a hard time finding GOOD pics of panaeolus mycelium, like with no glare. I don't care what kind, cyanescens whatever. Just would like it only 1/3-2/3 colonized across the plate. A cloned culture pic would be nice. :fishing:

P. foenisecii?

I've just transferred  plate of it and can post pics in a few days.  Somehow I figure it's not what you're after....

Unfortunately, I don't have any pans.

Here's a scanning electron micrograph of a gill section of coplandia.  You can see the four spores forming on each basidium, and a plurocystidia.  I know it isn't a petri dish picture, but those are all on another hard drive.

Pan cyan(copelandia) mycelium can resemble mold in its early stages.  Later, it's velvety white, but not rhizomorphic.

Thank you all for your efforts. You are all great.

This is my comparison. It is from the panaeolus I found posted in the hunting forum from So Cal. Thought to be pan. tropicalis

I am on the second transfer and am isolating more colonies tonight, probably should have last night though. Anyways. These are of the samples I sent to workman for microscopy.

Germination occurred on MYA in 23 hours on the second run. First run was multi spore streak from loop. To many colonies so decided to go with dilution. Dilution was around 1/100. (I know! It looks like there was to many colonies for that dilution, It was hard to not pick up so many spores from my print).  I did not gauge the first run but I think it was faster, due to heavy spore load. Growth occurs at a rate of about 2 - 2.5 mm per 12 hours and increased from a rate of 1 -1.5 mm for the first 1-2 days from inoculation. Contaminants were not too much of an issue and were fairly low for a wild print. Prints were also made on a place to place transfer on construction paper every 6 hours. Prints were dense and plentiful.

The top dish shows some sectoring mycelium, but the bottom dish is bacteria colonies.

Yes and no . Those might look like bacterial colonies ,but they are germinating spores "I promise". They previously looked like that when I made my first transfers. Yeah I though they were the first time around, but they are not. I wish I had a scope to show you up close. Maybe in the future. I know, the pictures make them look kinda sticky looking. It's because I let the water on the plate move over them.

Just plain. I don't bother with the stuff. Too $'y. You can't use the stuff when germinating spores anyways.

I've yet to ever use it.  Frankly, I've had good enough success without it by just using a few extra plates/samples.  I'm surprised that you find it too expensive though.  Which antibiotic do you find to be so expensive?

I germinate spores on antibiotic agar all the time.  It's preferred in fact.  If those are mycelium colonies, the picture must be out of focus.

Wow! how interesting. I never new that. I always thought it would hamper germination. Nice to know you can. I most often use charcoal agar for germination, this time I had a good 60 dishes already made up so I figured I better use them. I have never had the urge to buy some antibiotic solution. I have managed without it. thanks for the tip though.Of the many years that I have been doing this stuff. Granted the first 5 years were with cubes in po-dunk Oklahoma. I have never ever bought one bit of that stuff, because I never found a need for it and assumed it wouldn't work with germinating spores. Knowing that now, I can maybe use it knowing the price is worth it.  I have a few prints that are dirty as hell and are such a pain in the ass, I find myself not even wanting to bother.

Yeah! not trying to say you don't know your bacteria and all. I am sure you do. I am pretty familiar with them as well.  I have more plates that exhibit the same characteristics most indefinitely. I have subcultured from them and had no problems with it. It is definitely new to me  thats for sure. They seem to germinate into a fuzzy ball like mass that gets areal  and then starts to move across the plate. When they get wet it looks like your classic bacterial colonies until the culture grows out more.

I tried to get better pics this morning. I chose a different plate to use for transfer because of there being to many colonies .  Hope these are better for ya all. Look like pan mycelium?

Again i am thankful for your guy's input. And I got some antibiotic solution to try for the first time out of it. Thanks MycoAu!
