Liquid mycelium

My friend had a pint jar of liquid mycelium (the malt extract solution). He innoculated some jars w. it and within a couple days showed some great growth. About 4 days later he knocked up another batch, they didn't grow as strong or as fast. Another 3 or 4 days later, he innoculated another batch, and they have shown no growth at all. Any idea what could have caused this?? The jar wasn't kept in the fridge like it shoulda been, it just stayed in the incubator after it was already colonized. Could that have been the problem? Thanks!!!

...that might have been it, Visigoth...

I had some malt water that was kept in the fridge for 1.5 years and it was still good, so maybe it was the heat...

anyone else know??

Ok, glad u said that, because that leads right into my next question. Should I have injected more spores into the solution??? I only used 2 cc's, and even if 2 cc's is sufficient, the more spores u inject, the more mycelium you'll get in the solution because more spores will germinate correct?? I'm just thinking if I have to really fish for the mycelium in the syringes, then I should do something to allow there to be MORE mycelium in the solution. Any feedback or pointers??

more spores wont really make a difference. it might speed up the initial germination if you have it in a very large jar, but once the mycelium starts to grow it multiplies quickly

When you make your solution the mycelium will grow and spread.
So any amount of mycelium you use will spread.
I`ve kept a jar of honey mycelium in incubation for 2 months without a problem , put it in the frisge for 5 months and still Pefect.
So I think you might have another problem you haven`t addressed.
I would think nutrients have run out but the moment you inject the solution into a jra you are giving it more nutrients.
So you might have killed the mycelium with overly high temperatures.

The longer it incubates,the more all those little threads start to grow together in bigger structures,which starts to defeat the purpose.

Thanks guys, I have learned a lot from your replies. One more question, is there a noticeable difference between colonization if you innoculate a new jar of liquid w. mycelium rather than spores. Thanks again!!!!

Yes. You will see things go faster because you don't have to wait for the spores to germinate.

as you draw off living mycelium from your innoculant, what is left behind often has only a few clusters of mycelium to generate more from. there is a limit to how many generations a cell can copy itself before the whole mess becomes senesant, or basicly if you take a xerox copy of something that has allready been copied, and continue making copies of copies and not the original, eventually all you have is a blur. same thing happens in your licuid innoculant.

also, when you start a jar of licuid, it has no waste products, but after successive uses as described, any waste products will tend to build up in the licuid.

also, it is usual to have a certain amount of contam in licuid innoculant. a small amount goes unnoticed, but the contam is left in the jar to compete with your mycelium, and they have a competitive edge, since you were not trying to get the contams into your innoculation syringe. they all get left in the jar to compete with a depleated population of mycelium, which gets progressivly weaker as time goes by.

when i'm going to the trouble of making up a batch of licuid innoculant, i always make a few extra jars, and i innoculate from several, so that if one is contamned, i havn't wasted th whole works. i keep track of which jar the innoculant came from and if it does bad, i then know to throw it out instead of wasting more sterilized material and time. it requires little extra effort to make three jars as it does to make one.
