Four days ago i swabbed two petri dishes with Pan subhanburi spores from a print labeled 1/08... so now I'm looking at them and nothing... i have never had to wait 4 days to see growth on a dish before(usually if no myc then a contam of some sort), four days seems like a while, dose this happen? should i have hydrated the spores ?. I am only trying to get clean growth on the plate not an isolate i have read isolates from pans are hard to work with and many people don't get one they are happy with. after i get a decent amount of growth should i mix sterile water up in the petri to make some myco laden water or should i cut chunks out and put them in my jars ? which way would be the least contam resistant ?

yeah, i've had plates inoculated with dried spores take over a week to show. these days i follow germination at 640x or so with the scope.

If your sterile it doesn't which way you go. I like LC but I also make some petri dishes to use in jars if I want a good isolation for the LC. It is a little slower for RGS jars but for BRF cakes, it's like magic water.
