New petri dishes
    #8173065 - 03/20/08 06:42 PM (16 years, 30 days ago)

Do brand new petri dishes need to undergo any sterilization procedures before use, or is it good to pore the agar right into them as i take them out of the bag they are in.

Or some vendors sell dishes that are sterile until you open the packaging...those also need not be sterilized...if they are already sterile, they will say so on the packaging

They are sterile.

Unless they are already used they all come sterile.

Here's how to do it. Get a whiskey or wine bottle with a screw on lid that will fit in your PC. Drill a hole in the lid and stuff it with polyfill or cut a synthetic filter disk to fit inside the lid. Prepare your agar and pc in the normal way. The filter will prevent contaminants from entering the agar as it cools.
Build a glovebox. You can make one for the price of a clear rubbermaid. All you need is a couple of arm holes. You don't even need attached gloves, but if not, then be sure to wear latex gloves when you pour your dishes.
Allow the agar to cool almost to room temperature without opening the filtered whiskey bottle it's in. When it is cooled off, but still liquid is when you want to open it up to pour. Work fast. Open your sleeve of presterilized dishes just before use. Wash the outside of the sleeve with alcohol before opening carefully from one end.
Stack your dishes in two piles of ten. pour the bottom dish by lifting the entire stack, then set it down and pick up the other nine. Repeat until you've filled both stacks, then CAREFULLY insert the sleeve back over the petri dishes and seal the bottom so it's airtight.
The above procedure should give you a 100% success ratio with a bit of practice. I find no difference in contamination rates with a glovebox or flowhood, but of course the flowhood gives you more elbow room and is much easier to work in front of.

I just got 300 plastic petri dishes that can be PCd !
unbreakable and reusable polypropylene.
Designed for microwave sterilization in labs.
perfect size too 50 mm.
20 for $25 if you PM me (I pay shipping in Canada), I don't need them all but since the shipping was the same on 300 as 30 I bought a case.
Nice to work with, drop a chunk of agar in it PC it and let it cool...good to go!

Yeah but yours melt if you cook em and crack if you squish em.Probably polystyrene. You may be able to re use them if you soak them in bleach or alcohol though.
PP is a compromise between glass and disposable plastic.
I got some cultures in the mail in brittle plastic dishes once, half of them cracked, lost some cool cultures that trade.I've been looking for something better since then.
It is pricey but you get the best qualities of Pyrex and polystyrene in one petri dish.

what does PP stand for? i've been looking for some re-usale (PCable) plastic petri dishes, since the glass ones are outragously expensive!

PP means polypropylene same stuff spawn bags are made of.
and these!
These are glad PP kitchen containers, I'm using them for cakes, nice taper and a torch sterilized needle goes right thru the lid without drilling a hole. Tape it right up afterwards of course.

I had to order from a medical supply company to get my PP dishes, you won't find them at mushroom supply places.
I used to use glass but yeah it really costs you and shipping can be iffy.
PP is a good compromise and I can fill them and then PC them instead of screwing with a flow hood or glove box and trying to do a sterile pour with hot agar.
