Sterilising petri dishes?

Hey everyone, Just a quick question here. A couple of days ago I poured 9 petri dishes with some agar and it didn't set - (oops - still experimenting with ratios) anyway, I only have like 15 clean petris left so I was wondering what is the best way of sterilising these for re-use? They are the standard plastic kind. Alcohol wipe? Boiling?

Never tried to work with used plastic dishes. If it were me, I would throw them away.

They are not ment to be reused. But IF, I?d recomend you to wash them thorouhghly with bleach water, let dry and wipe with alcohol or peroxide.

I'm not doubting your advice here Anno. I'm just curious to know if you've tried this in the past and what the failure rate was.

I tried this, yes. but only on 5 dishes. I was using a flow hood and 1 dish contaminated. I used iso-propanol, 70% to wipe the dishes.

Thanks for the advice guys. I'll give it a try. Otherwise I'll just get some more petri dishes. It's not that difficult, it just involves acting like I'm doing something legitimate to some scientists....or is there an easier way of getting petris?

I just mail order the reusable glass kind. If you can't do this and you don't want to have to acquire more locally, you have alternatives. Before I had access to petri dishes, I would use 1/2 pint jars (the standard PF jar) with a bit of agar in the bottom. Just PC the jars after adding the agar to sterilize them. Another alternative would be to tell the scientists who give you the dishes that you are cultivating oyster mushrooms and need to play with agar so that you can create spawn.

why bother ther 4-5 euro for twenty or buy a box anter like 2-3 for twenty and yes you grow mushrooms sow what thats legaly bistores can only be started that way ask for pda while your at it its cheap and good quality

please don't take this to be a snyde remark of a jackass, but has it occured to anyone that there isn't a huge differance between a petri-dish and a wide mouth half-pint mason jar?
i use them with resealable plastic lids for many purposes like small culture stuff, storage of spore-print taken directly inside the jar, etc. both the jars and the lids are pc safe.
the biggest advantage of using an ordinary jar is that when you are done, you can still use the jar for other things, while a petri dish can only be used as a petri-dish. what's more, the plastic lids do a reasonable job of sealing against outside airborne contams, petridishes have to be taped.

I've had success re-using polystyrene petris by washing with soap and water then boiling for 5 mins. But then, I use peroxidated agar all the time, so sterility isn't so important.

What would happen if you had a half pint w. agar in the bottom and u layed a fresh cap on top of the agar and let all the spores fall onto it, that would be huge overkill huh?

I'd rather peer into a petri dish when looking at the contents as opposed to trying to look into a jar which has an opaque lid and must be viewed from an acute angle.

Uhm, how is your post relevant to the question he asked? I believe he was asking about multi-spore innoculation of agar and not cloning. Please read his question again for clarity if you don't understand:

I figured why multi-spore when you can just clone a fruit you liked. I am sorry if this rubbed you the wrong way man, but I would rather do that than multispore. Figured I could suggest something. More even flushing, no wondering about what kind of fruits you will get, etc.. entheo

Not too mention that mushrooms in general seem to senesces relatively rapidly from cloning too often. I am sure you could clone for a few crops, but after that you will probably start noticing strain degeneration. With multi-spore work you have the advantage of genetic diversity. I think both are good, but I consider clone work to be more of a temporary way to simply things. (In terms of homogeneity of the fruit. As far as petri dishes go... I recommend getting glass ones. Environmentally friendly, re-usable, autoclavable... wonderful
