Tricks for faster wbs colonization
    #8036531 - 02/18/08 02:32 AM (15 years, 11 months ago)

I have seen posts here where people have fully colonized quart WBS jars in under two weeks. What are your secrets?

I run water through my seed to clean it, take out about 3/4ths of the sunflower seeds, let sit for 15+ hours, run water through until clear and then drain until the right water content is reached. Sometimes I add verm if its too wet. I use a series of small holes in the jar lid covered by tyvek and inoculate with anywhere from 3-10cc's healthy LC.

It takes me 20+ days in an incubator to get 100%

What can I do to speed this up? I have been experimenting with coffee, gypsum and hydrated lime, not too much noticeable difference.

What do you two weekers do to speed up your jars? Does a good isolate speed up growth significantly? Thanks.

Some have had success with olive oil, but I would consider that an experimental technique.

After your initial rinse brew a pot of weak coffee and pour it in your WBS. Add enough eater to cover the seed and let it soak. If you are using multi-spore that will add 5-7 days to colonization. I use LCs to inoculate my WBS and along with the coffee soak I usually have full colonization in about 6 days.

I just started to soak in coffee and am going to see how that goes. My last batch of jars stalled because of too high temps but normally I can get near 100% colonization in 2 weeks.

I will let the seed sit in hot water (out of the tap no need to boil) and scrap any floaters off. After about 30 min - 1 hr I'll rinse if off and put it back in hot water. Let it sit for 6-12 hours, rinse add new water. Soak for another 6-12 hours. Then I'll rinse one more time and put the WBS on to boil. I let it get to boiling and then turn it off and stir it REALLY well. I let about 70% of kernals bust (about 4-5 hours) but most would say that 20% is the max you want (it will release starch which slows colonization.

Since mine are over saturated with so many burst kernals I will just let it drain off completely for an hour or so. Then I'll put it in jars with 1 tbs of gypsum (it just helps with shaking later). I'll give it a really good shake before I PC. Once the PC is done and back to normal pressure I will take the jars out and give them another REALLY good shake (use pot holders). That helps the seeds stay seperated more than anything.

I do the same as you with the holes and healthy LC but only about 3-4 CC, more than that is too much water. When it is incubating I will rotate the jars, top to bottom, inside to outside of the incubator. That way you prevent all the direct heat from the sides/bottom from slowing your growth.

Just let the jars sit, don't constantly shake. When there are a few really healthy myc spots I will gently roll the jar. I don't shake it, just rolls it so they come into contact with the neighboring kernals. At 25% or so I will break up the myc chunks by shaking and try and get them positioned in non-colonized areas.

After that I don't touch them until they are 100%.

My quart jars of WBS colonize fully in 6 days:shocked:(Some take 7 or 8 days, but, most take only 6 days)

Doc's WBS Tek + LC + 80*F + strain + shaking at the right time = 6 day colonization times!

Need I say more:cool:

you can also do a a little work with ur jars to find the fast colonizer. say you knocked up 5 jars and one of them colonized in 13 to 14 days. use that one for ur g2g transfer... if u do a g2g from 1 to 10 jars... u are looking at about 6 to 7 days for 100% colonization of 10 jars.

also recommend shaking after spore inoculation and g2g

Definitely agree with Doc on this one. Inoculating with a liquid culture, the right temps, shaking around 20-30%, and strain will always yield the fastest colonization times. Spores, due to the delay in germination, almost always take 2-3 weeks.

Step up to LC and you'll see the difference immediately.

totally agree with Doc and Wronguy

I've tried soaking in coffee but full colonization still takes about 6 or 7 days regardless, assuming I use LC of course

When you guys inject the LC do you squirt it in one spot against the glass or do you rotate it around? Do you shake right after inoculation?

I have had wbs jars colonize in under two weeks with LC. LC is the key to fast colonization.

No doubt, looking for other factors though.

I have 3 pints of wbs colonizing now. 1 is almost 100% at its 14th day since multispore inoc. Later today I plan on sterilizing up to 10 Quarts of WBS to do G2G from pint to Quarts in hopes to have up to 10 quarts of WBS to case...

I have a question about the coffee, what role does it play in "faster colonization"?

if you're doing G2G that is the absolute fastest way to inoculate a jar

in regards to the coffee, i find that it doesnt make a significant difference in colonization times if you are using LC, if using MS it would likely speed up colonization by at least a couple of days

the reason I use coffee is for the extra nutes when spawning and fruiting, im honestly not sure if it makes much of a difference but being a big coffee drinker i usually have half a pot left over from the morning so i just toss it in when i soak the grain

you can colonize quarts in roughly 3 days tops if you use something similar to 9er tek and have one pf cake available. very simple, and its amazing. one pf cake can become at least 50-70 quarts of liquid inoculum(sp? lol_).

sorry rephrase this. one pf cake can make enough liquid culture to colonize 50-70 quarts in 3 days tops! and that is a fact, ive done this method 3 times with 100% success.

the one thing you have to watch out for, is if you steep your grain. you have a bit more moisture in the jars. you can either soak up the excess moisture that will be added when you pour lc into your jar by adding dry verm. or use less solution. shake very well. and it works like clockwork!

look up 9er tek. i think it should be somewhere in the archives here. don't know these forums well.

Yeah, I need to find a way to speed up colonization time. I inoculated a bunch of jars on the 4th. They were about 1/4 way done and I shook them. Now they look completely colonized, but the mycelium is very thin. Almost reminds me of cobweb. I don't think it's cobweb because it doesnt fluff up at the top or anything. I'm going to give them a few more days to see if the mycelium thickens up. Liquid cultures don't seem to work too well for me.

I love the posters who show up every time, in every old thread that gets bumped to call out the other posters for bumping the thread.

This thread answered 2 questions for me.  1, if an opt-out can become a Trusted Cultivator and 2,  There's doc34. doc34 is the originator of the infamous, outdated, Doc's WBS tek.  Not our other house doctor.

To me, old threads being bumped are evidence that the search function is still being used by new members.

Any thread 2+ yrs IMHO is suspect let the dead threads remain dead.  This thread has absolutely no substance to it what so ever.  So what are you really trying to say to me dr innoc.

Edit nvm
