Hello all.

Some weeks ago, i managed to clone an Oyster shroomie succesfully, but i failed afterwards, probably because i didnt know *EXACTLY* what to do with the substrate to transfer it to. I made something of my own and as it was expected it didnt worked.

So, now i am gonna try to move a cloned mycelia of Agaricus (portobello actually) to some kind of substrate. I know that Stamets is the Master, but my pocket holds the Master of Emptiness so i have to wait for some months till i manage to buy his great books. Can somebody be so kind to post what is the (a) exact substrate and (b) exact tek that stamets write about Agaricus? There is no need to ocr the whole chapter [although it would be soooooo f**** cool! ] but just write down what the ingredients and quantity are and how to prepare the substrate in a few but clear words.

Has anyone succeded with Agaricus in this forum? I tried and tried searching but no pics of succesfully home-cultivated of Agaricus had i found... But again, it may be that i am a fool, so if u know any forum-links, i would much appreciate it!

Clone on potato-dextrose-yeast agar.....transfer to rye grain.....case with a deep layer of un-steralized compost.

hm...

about this transfer: how is it possible to transfer it to lets say a jar of sterilized rye grain, without jeopardizing the whole sterility issue? I suppose it has to be done with a syringe. But if i pull the mycelium into a syringe from the cloning jar, wouldnt that hurt it? mycelium has parts that are just white and parts that are like roots, so if mixing them alltogether, wouldnt that confuse or even destroy its ability to colonize?

Pulling mycelium into a syringe won't hurt it, the only thing you have to worry about is sterility. It's better to transfer part with "roots" (rhizomorphes) but IMO mixing wouldn't hurt its ability to colonize.
You can increase the chances by spraying few ml of H2O2 in the grain jar before inoculation. It keeps contaminants away and doesn't hurt the mycellium. I had very poor results without it.

I am also a "fan" of hydrogen peroxide, so its okee. well, lets try to solve another similar puzzle to me: lets say that i have a jar of healthy colonized mycelium. Now i want to transfer some part of it into other jars, for faster colonization, as i have already read in some teks. Unfortunately, i dont quite understand how am i supposed to transfer a piece of mycelium cut with a sterile sort of knife into some rye grain jar. How am i supposed to keep the rye grains sterile after the pc, as i have to open the jar and throw the mycelium inside? I dont think i can use a syringe to transfer colonized rye grains Even using h2o2, bacteria will definately have a party in there, starting their invasion as soon as i am opening the jar.
Do you think that if i am fast and work inside the warm oven so as to create the famous "thermal mushroom" to protect the job is enough?

I don't have a flow hood and a glove box. I clean all surfaces in my "lab" with bleach before inoculations, I also wipe external parts of jars before opening. I cut wedges of mycellium with flame sterilized scalpel, open grain jar, spray H2O2 inside and put in few wedges of mycellium. IMO the most important thing in preventing conamination is the right moisture content of grain and proper sterilization. If your grain is too moist and has many exploded kernels then peroxide will not help.
About 10% of jars get contaminated. I expect that with a flow hood the contamination percentage would get close to 0%.

kykeon: you'll probably be fine.....uses dilute bleach or iso alcohol to wipe down surfaces, including the outside of jars you're handling. Flame treat your scalpel/innoculation tool and then wipe them with iso alcohol too.

Yeah; When you start doing transfers it's time to build a glove box. I'm building one now but mine will be open where the two holes for your hands would normally be
and two strong zerox copier fans will be pushing air through a 99.9% hepa filter
mounted on the top/ rear of the glove box. So it's kind of a glove box/ flow hood.
I ripped this design off BTW . I can't remember where. Mycotopia has a good design for a flow hood. I'm doing my glove box this way because i have all the stuff I need laying around my garage
